Objective: To observe the expression of miR-451 in placental tissue and peripheral serum of patients with preeclampsia,and the expression of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)in placental tissue The relationship between miR-451 expression levels and MMP-2 and MMP-9 expression levels was analyzed to provide a research basis for the study of the mechanism of preeclampsia and provide a clinical basis for early monitoring of preeclampsia.Methods: With the approval of the Medical Ethics Committee of Medical Ethics Committee of Jining Rencheng Maternal and Child health Hospital,30 cases of early-onset preeclampsia patients,30 cases of late-preeclampsia patients,30 cases of premature pregnant women,and 30 cases of normal pregnant women were selected from December 2018 to May 2019 as research target,which set as early-onset group,late-onset group,premature group and healthy control group.All enrolled pregnant women were drawn with venous blood before delivery,serum was separated,and the expression level of miR-451 in peripheral serum was detected by real-time quantitative PCR.At the same time,placental tissues were collected during delivery,and real-time quantitative PCR was used to detect miR-451,MMP-2 mRNA,and MMP-9 mRNA expression levels.Western Blot techniques were used to detect the expression of MMP-2 and MMP-9 protein in placenta.SPSS statistical software was used to analyze the relationship between the expression levels of miR-451 in placental tissues and peripheral serum of pregnant women in each group and the expression levels of MMP-2 and MMP-9 in placental tissues.Results:(1)There were differences in the expression levels of miR-451 in peripheral serum and placental tissue of pregnant women in each group,and the differences were statistically significant(P <0.05).The expression level of miR-451 was highest in the preeclampsia group of pregnant women,followed by the late-eclampsia group,followed by the premature mother group,and the normal pregnant group was the lowest.The difference between the groups was statistically significant(P <0.05).(2)There were differences in the expression levels of MMP-2 mRNA and MMP-9mRNA in pregnant women in each group,and the differences were statistically significant(P <0.05).The expression levels of MMP-2 mRNA and MMP-9 mRNA in pregnantwomen in the pre-eclampsia group were the lowest,followed by those in the late-eclampsia group,followed by the premature mother group,which was highest in the normal pregnant woman group.The differences between the groups were statistically significant(P <0.05).(3)There were differences in the expression levels of MMP-2 and MMP-9 proteins in pregnant women in each group,and the differences were statistically significant(P <0.05).The expression levels of MMP-2 and MMP-9 proteins were lowest in pregnant women in the preeclampsia group,followed by those in the late-eclampsia group,followed by the premature mother group,which was highest in the normal pregnant woman group.The differences between groups were statistically significant(P <0.05).(4)The expression levels of miR-451 in the peripheral serum of pregnant women in each group were negatively correlated with the expression levels of MMP-2 mRNA,MMP-9m RNA,MMP-2 protein,and MMP-9 protein in placental tissues(P <0.05).(5)The expression levels of miR-451 in placental tissues of pregnant women in each group were negatively correlated with the expression levels of MMP-2 mRNA,MMP-9m RNA,MMP-2 protein,and MMP-9 protein in placental tissues(P <0.05).Conclusion: The expression levels of miR-451 in peripheral serum and placental tissue of preeclampsia pregnant women are higher than those in preterm and normal pregnant women.Among them,the expression levels of miR-451 in peripheral serum and placental tissue of pregnant women with preeclampsia are most obvious.In addition,MMP-2m RNA,MMP-9 mRNA,MMP-2 protein,and MMP-9 protein are also lowly expressed in placental tissues of preeclampsia pregnant women.This study shows that the indicators of preeclampsia women also changed most obviously.The expression levels of miR-451 in peripheral serum and placental tissue and MMP-2 mRNA,MMP-9 mRNA,MMP-2protein and MMP-9 protein were negatively correlated.
