ObjectiveThe purpose of this paper is to study the impact of nattokinase on sclerotin of rat with osteoporosis caused by alcohol.MethodThe method was to select 60 2-month-old male rats and,based on their weight,randomly divide them into four groups: normal group,alcohol model group,nattokinase control group,and nattokinase treatment group.The normal group were given normal saline once a day.The alcohol model group were given 8ml of 50%(V/V)alcohol every day for two weeks,and 12ml/(kg·bw·d)for the next six weeks.The nattokinase control group were given 530.39(fu/g)of nattokinase capsule solution every day.The nattokinase treatment group were given 530.39(fu/g)of nattokinase capsule solution and the same amount of alcohol as the alcohol model group for eight weeks.Use ICP-AES to detect the level of blood Ca and P,ELISA to detect the level of BGP and BMP-2,H&E staining to observe the pathologic change of the rat femurs,DEXA to detect BMD of the rat femurs.The following results were obtained: Result1.The alcohol model group showed a significantly lower level of BMD than the normal group(P <0.05),and BMD level of the nattokinase treatment group was obviously higher than the alcohol model group(P <0.05);2.The H&E staining results showed that bone trabecula of the normal group was dense,regular and thick,with an even thickness;bone trabecula of the alcohol model group was discrete,irregular,with an uneven thickness and even fractures;bone trabecula of the nattokinase treatment group was dense,regular and thick,with an even thickness,no fracture was found;3.Blood serum test results showed that the level of blood BGP,BMP-2,Ca and P of the alcohol model group were significantly lower than the normal group(P <0.05);the nattokinase control group and normal group showed similar blood serum rest results;the level of blood BGP,Ca and P of the nattokinase treatment group were significantly higher than the alcohol model group((P <0.05).4.The results of serum endotoxin indicators showed that: Compared with the control group,there was no significant change in nattokinase control group,the level of serum endotoxin in model group increased significantly(P<0.05).Compared with model group,the level of serum endotoxin decreased significantly in Nattokinase intervention group(P<0.05).ConclusionThe study led to the following conclusion: rat with AOP suffers from significantly decreased bone formation and bone mineral loss;Nattokinase can help increase bone formation of rat with AOP and restrain bone mineral loss and has an obvious protective effect on osteoporosis treatment inrat.
