| Objective:1.Through the observation of 24-hour urine protein quantity,serum albumin,renal pathological changes,to explore the therapeutic effect of QDGS formula and its decoctions on IMN rats,and to determine the contribution of each decoctions group in the whole formula;2.By observing the effect of QDGS recipe on the activation of NF-?B in renal tissue of IMN rats,the mechanism of QDGS recipe in the treatment of IMN was further clarified.Method:In this study,we used cationic bovine serum albumin(C-BSA)to duplicate the rat model of IMN.65 male SD rats of SPF grade were randomly divided into 6 groups: Ten blank groups were reserved.After the model was made successfully,they were divided into five groups: model group,benazepril group,QDGS Group(hereinafter referred to as TCM Quan Fang Group),PBGS group and QLHX group.The blank group and the model group were given the same amount of saline for 28 days.The levels of 24-hour urine protein,serum albumin,total protein,triglyceride,total cholesterol,transaminase,urea nitrogen and serum creatinine were observed,and the pathological changes of kidney.To observe the expression of NF-?B activation related protein in kidney tissue of blank group,model group,benazepril group and traditional Chinese medicine group.Result:1.QDGS and its decoctions can reduce the 24-hour urine protein content of IMN rats.The 24-hour urine protein quantity of all groups was lower than that of the model(P<0.05),the TCD and Peibu Gushen group is quite remarkable than the model(P<0.01),the TCD group is quite remarkable than benazepril(P<0.01),There was no difference between the two groups(P > 0.05).2.QDGS and its decoctions can increase serum albumin and total protein of IMN rats.The level of TP in each group was higher than that in the model group(P<0.05),The TCD group was higher than benazepril group(P < 0.05),There was no difference between the two groups(P > 0.05),other results of total cholesterol analysis were synchronous with those of triglycerides.3.QDGS and its prescriptions can regulate the lipid metabolism of IMN rats.The TC of all group was significantly lower than that of model group(P < 0.01),TC of TCD group and PBGS was lower than that of benazepril(P<0.05),TC of PBGS was lower than that ofQLHX(P<0.05);TG of TCD group was significantly lower than that of model(P<0.01),TG of TCD group was lower than benazepril group(P< 0.05).4.There was no significant difference in ALT and AST between all groups(P>0.05).5.QDGS can reduce the renal pathological damage in IMN rats.QDGS decoction can reduce the renal pathological damage to some extent.6.ELISA prompts QDGS can reduce the expression of IL-1?,TNF-?.The expression of IL-1? and TNF-? in benazepril group and TCD group was significantly lower than that in model group(P < 0.01),the IL-1? of TCD group was significantly lower than benazepril(P < 0.05),There was no difference between the TCD group and benazepril in TNF-?.(P >0.05).7.WB prompts QDGS can reduceand NF-?B p65 in renal tissue of IMN rats,and inhibit the degradation of I ? B ?.The results of NF-?B p65 and I ? B ? analysis were the same in each group,which showed that there was significant difference between each group and model group(P < 0.01),between benazepril group and traditional Chinese medicine group(P < 0.01),there was no difference between model group and benazepril group(P > 0.05).Conclusion:1.QDGS can reduce 24h-utp,increase ALB and TP,regulate lipid metabolism,alleviate renal pathological damage;2.The therapeutic effect of QDGS Decoction group was better than that of the whole variance group,and the therapeutic effect of Peibu Gushen Decoction group was better than that of Qingli Huoxue group,suggesting that QDGS Decoction group was scientific and rigorous,and Peibu Gu Shen medicine group played a major role in the prescription;3.QDGS can down regulate the expression of NF-?B p65,TNF-? and IL-1?,inhibit the degradation of I ? B ?,suggesting that qdgsr may reduce the renal injury of IMN rats by inhibiting NF-?B activation. |
