The Investigate About The Effect Of Zuogui Pill On Reproductive Function Of Rats With Excessive Weight Loss Was Based On The Expression Of Bcl-2/Bax

Objective:this experiment used the method of"l-carnitine+half diet+excessive exercise"to cause excessive weight loss in rats.Bcl-2/Bax in ovaries and serum sex hormone levels of rats at different stages were used as entry points to investigate the effect of excessive weight loss on reproductive function of rats.To investigate the effect of zuogui pill on reproductive function reversal and the correlation between the degree of excessive weight loss and zuogui pill intervention.Methods:100 rats with regular estrous cycle were selected for the experiment.20 rats were randomly selected as blank groups.60 rats adapted to treadmill training were selected and divided into 6 groups of 10 each.Group A was divided into A1,A2,A3 and A4,and group B was divided into B1,B2,B3 and B4,followed by blank group,model group,restoration diet group and restoration diet+zuogui pill group.Except for the blank group,all rats in group A were given"l-carnitine+half diet+running table exercise"in 1-4 weeks.Four weeks later,A1 and A2 were put to death to compare the differences of various indicators between the two groups.At 5-8 weeks,A3 and A4 resumed their diet,stopped l-carnitine and platform running,and at the same time,A4 gave zuogui pill to intragastric administration.After 4 weeks,A3 and A4 were killed to compare the differences of various indicators between the two groups.The modeling time of group B was 8 weeks,and the experimental method was the same as that of group A.Results:1.Changes in weight:the weight of rats decreased by 17%at 4 weeks and decreased 23%at 8 weeks,which were significantly different from the blank group?P<0.01?.2.Changes in the estrous cycle:the estrous cycle of the model rats at 4 and 8weeks was disturbed,which was significantly different from the blank group?P<0.01?.3.Changes of sex hormones:intra-group comparison:FSH and LH were increased at 4 and 8weeks after modeling,E₂ Compared with the blank group,there were significant differences?P<0.01?.After 4 weeks and 8 weeks,FSH,LH and E₂ decreased Compared with the modeling group,there were significant differences?P<0.01?.After 4 weeks of model making,there was a significant difference in FSH and LH decrease between the group receiving restoration diet+zuogui pill and the group receiving restoration diet only?P<0.01?,E₂ The increase was statistically significant?P<0.05?.After 8 weeks of modeling,there was no significant difference in the changes of relevant indicators between the group receiving the restoration diet+zuogui pill and the group receiving the restoration diet only?P>0.05?.Group comparison:FSH,LH,E₂ at 8 weeks of modeling₂There was significant difference?P<0.01?between the two levels at 4 weeks.4.Changes in the expression of Bcl-2/Bax in the ovary:intra-group comparison:compared with the blank group,there were significant differences in Bcl-2 decrease and Bax increase at week 4 and 8 of modeling?P<0.01?.Compared with the model group,4 weeks after modeling and 8 weeks after the resumption of diet,the increase of Bcl-2 was significantly different?P<0.01?,and the decrease of Bax was statistically significant?P<0.05?.Compared with the simple restoration diet group,the increase of Bcl-2 in the restoration diet+zuogui pill after 4weeks of modeling was statistically significant?P<0.05?and the decrease of Bax was significantly different?P<0.01?.There was no significant difference?P>0.05?between the model group+zuogui pill after 8 weeks of diet restoration and that of the pure diet restoration group?P>0.05?.Comparison between groups:the expression of Bcl-2/Bax in ovaries at 8 weeks was significantly different from that at 4 weeks?P<0.01?.Conclusion:1.The method of"l-carnitine+half diet+excessive exercise"can lead to the slimming state of rats.2.When the weight of the rats dropped by 17%,the ovarian function was already decreased.The resumption of diet could improve the reproductive function damage to some extent,and the effect of reversing the reproductive function damage was better when combined with zuogui pill.3.When the weight of the rats dropped by 23%,the ovarian function was worse,and even failure occurred.After given restoring diet and zuogui pill,the effect of reversing the damage of reproductive function were not good.
