Objective:The purpose of this study was to investigate the expression of matrix metalloproteinase11(MMP11)and disintegrin-metalloproteinase 28(ADAM28)in human breast cancer and the normal breast tissues adjacent to the cancer and its relationship with clinicopathological parameters.And the correlation between expression of the two proteins,analysis the effect what they are occurrence,invasion and metastasis in breast cancer,and the possibility of using it as a marker of breast cancer progression,it provides a theoretical basis for seeking new treatments with the breast cancer.Methods: Selected specimens of improved radical mastectomy for surgical breast cancer in the First Affiliated Hospital of Jiamusi University of Heilongjiang Province from November 2017 to May 2019.There were 80 cases of invasive ductal carcinoma confirmed by two senior pathologists without any antitumor treatment and have complete clinical data.This experiment was divided two groups,the one is experimental group and the other one is control group.The experiental group consisted of 80 cases of invasive ductal cancer paraffin tissue(40 cases of Her-2(3+)and 40 cases of Her-2(0)).The control group consisted of 30 cases of para-carcinoma normal breast paraffin tissue.Pass immunohistochemical staining(PV8000-Vision)was used to detect the expression of MMP11 and ADAM28 proteins in invasive ductal carcinoma and normal tissues adjacent to the carcinoma.Results:1.MMP11 and ADAM28 proteins are expressed in breast invasive ductal carcinoma tissues and adjacent normal tissues,and the positive expression rate of MMP11 protein in invasive ductal carcinoma tissues is 72.50%(58/80),in normal tissues adjacent to cancer The positive expression rate of MMP11 was 16.67%(5/30);the positive expression rate of ADAM28 in invasive ductal carcinoma tissue protein was 75.00%(60/80),and the positive expression rate in normal tissue adjacent to the cancer was 26.67%(8/30).The results showed that the expressions of MMP11 and ADAM28 proteins in cancer tissues were significantly higher than those in adjacent normal tissues,and the difference was statistically significant(P < 0.001).2.The positive rates of MMP11 proteins and ADAM28 proteins all increased with the increase of Her-2(3+),histological grade ?,and lymph node metastasis(P < 0.05);it was not related to the patient's age,tumor size,and PR expression(P > 0.05).However,in studies related to ER,it was found that the expression of MMP11 and ADAM28 are inconsistent.The positive expression of ADAM28 has nothing to do with ER(+).The ?2 value is 1.509,P > 0.05,and the positive expression rate of MMP11 in the ER(+)group is significantly higher than that of ER(-).The group difference was statistically significant,with a ?2 value of 12.122,(P < 0.001).3.Analysis of the expression of MMP11 protein and ADAM28 protein in the experimental group and their correlation: When the positive expression of the two was statistically analyzed,in 51 cases,the expression of MMP11 and ADAM28 in the tumor were both positive and negative There were 13 cases.There was a positive correlation between the expression of the two proteins in the experimental group(r = 0.485 P < 0.001).Conclusion:1.The expression of MMP11 and ADAM28 in breast infiltrating ductal carcinoma tissue was significantly higher than that in normal tissues adjacent to the cancer,indicating that both MMP11 and ADAM28 are involved in the occurrence of breast infiltrating ductal carcinoma.2.The positive expression of MMP11 and ADAM28 protein is related to Her-2(3+),histological grade III and lymph node metastasis,but not to the patient's age,tumor size and PR expression,suggesting that the high expression of the two may be related to The progression and invasiveness of invasive ductal carcinoma of the breast are closely related.3.Positive correlation between MMP11 and ADAM28 protein expression in breast invasive ductal carcinoma.
