Objective:In order to understand the serum infection rate of KSHV in the normal population in Shihezi,Xinjiang,a study was conducted on the infection status of KSHV in the general population in the Dead Shihezi,Xinjiang.It also analyzed the risk factors of KSHV infection,and provided a relevant molecular biological analysis for the continuation of the epidemiological investigation of KSHV in China.It provided a scientific basis for the local health authorities to prevent and block the infection and transmission of KSHV.Methods: The prokaryotic expression vector p QE-80 L and E.coli BL21(DE 3)strains were used to express His recombinant virus proteins ORF 65,ORF 73,and ORF K8.1?.Three recombinant proteins ORF 65,ORF 73,and ORF K8.1? were obtained by Ni2 + affinity chromatography.The immunogenicity of the recombinant protein was detected by Western blot.Standard albumin was used to draw a standard curve for the final protein concentration of Bradford method,and the final concentrations of three recombinant proteins were finally measured.After knowing the final concentrations of the three recombinant proteins,the recombinant protein was used as the antigen,and the optimal conditions for the antigen-antibody reaction were obtained using a chessboard method,and a complete ELISA indirect method for measuring KSHV antibodies in serum was established.Using the established ELISA indirect detection system,more than 1,000 ordinary human serum samples were investigated in Shihezi,Xinjiang,and the test results were obtained.And use SPSS software to analyze and process the detection results.Results: The total positive rate of serum antibodies for KSHV infection in the general population in Xinjiang was 25.60%.Logistic univariate regression analysis of the test results found that there was no statistical difference in the gender of the infected population.The male infection rate was 22.86% and the female infection rate is 25.54%.However,among the famous ethnic groups,the infection rate of Uighur ethnic group was 29.79%,while that of Han ethnic group was only 9.69%,which was statistically different(P <0.001).There are also differences in occupations.Among the agricultural and pastoral populations mainly in agriculture and animal husbandry,the KSHV serum infection rate is 27.70%,and the KSHV serum infection rate in other populations including workers and cadres is 19.29%.The difference between them was statistically significant(P = 0.002).In terms of age distribution,the serum infection rate of KSHV in people older than 50 years old was 20.18%,and the serum infection rate of KSHV in people less than 50 years old was 27.48%.In the exercise frequency of the surveyed population,the analysis found that the serum infection rate of KSHV for regular exercise was 16.56%,while the serum infection rate of KSHV for those who did not participate in exercise was 25.52%.The difference between the two was statistically significant(P = 0.018).Statistical analysis found that other factors,including education,BMI,smoking,and drinking,had no statistical significance for KSHV infection(P> 0.05).Logistic multivariate regression analysis of these factors found that there was a significant correlation between KSHV infection and KSHV infection among Uighur ethnic groups,vegetarian diets,and vegetarian diets(P <0.05).Conclusion:The KSHV infection rate is higher among the general population in Shihezi,Xinjiang,northwestern China,and there is no significant difference in gender between KSHV infections.However,there were significant correlations between KSHV infection and factors such as clan,occupation,age and frequency of exercise.
