Objective This study aimed investigate the association of CAV-1 polymorphism with breast cancer in Chinese female populationof Gansu Province.Methods A total of 200 cases of female breast lesions with pathologically by fine needle aspiration biopsy and / or surgical resection were selected as the experimental group,consist of 52 benign and 148 malignant lesions,at the same time,166 healthy women were selected as healthy control group.Kompetitive Allele Specific PCR(KASP)was used to analyse the genotype of CAV-1 gene polymorphims.Spss 22.0 software was used to analyze whether the selected cav-1 gene conform to the Hardy-Weinberg genetic balance,and if so,Chi-square was analysed to compare CAV-1 genotypes and alleles in different comparative groups(Benign/Healthgroup,Malignan/Health group,and Malignan/Benign group).The correlation between genetic polymorphism with susceptibility of benign and malignant breast tumor were analysised by binary logistic regression,the relationship between CAV-1 gene polymorphisms and clinical and pathological characteristics of BC group was also analyzed by the same method.Utilizing the Haploview to analyze the linkage disequilibrium between rs3807987 and rs7804372,and the relationship between the susceptibility of BC and haplotypes,which was constructed by rs3807987 and rs7804372 were anslysised by the shesis online software.Result(1)The CAV-1 SNP sites we selected were accord with Hardy-Weinberg genetic balance.(2)There was no significant difference in the frequency distribution of rs12672038 genotype and allele among the comparison groups;The distribution of rs3807987,and rs7804372 genotype and allele were significant difference in malignant/healthy group,but no statistical difference in benign/healthy,and malignant/benign groups.(3)The relationship between CAV-1 gene rs3807987 ?rs7804372,and rs12672038 with the risk of breast benign lesions were not found,and rs12672038 with the risk of breast cancer was also no correlation;There were significant association of rs3807987 and rs7804372 with risk of breast cancer were found in codominant model,dominant model and allelic model.The risk of breast cancer was increased with rs3807987 AG or AG/AA genotypes compared to those with GG genotype,and increased with A compared to G allele;The risk of breast cancer was increased with rs7804372 AT or AT/AA genotypes compared to those with TT genotype and increased with A compared to T allele.(4)There were no significant association of CAV-1 rs3807987 and rs7804372 polymorphisms with axillary lymph node metastasis,ER,and Ki67 protein expression.(5)Strong Linkage disequilibrium was found between CAV-1 rs3807987 and rs7804372,but no association between the haplotypes with the risk of breast cancer.Conclusion There were no relationship among CAV-1 gene rs3807987?rs7804372,and rs12672038 with the risk of breast benign lesions.The correlation between rs12672038 and the risk of breast cancer was also not exist;CAV-1 rs3807987 and rs7804372 were associated with susceptibility to breast cancer,carrying rs3807987 A allele?(GG?AG/AA)genotypes or rs7804372 A allele?(TT?AT/AA)genotypes increased the risk of breast cancer.
