Objective:To explore the correlation between the polymorphism of ER?gene Xba?and Pvu?and the susceptibility of breast cancer with its four molecular subtypes based on famale.Methods:This study sequentially collected 108 cases of female breast cancer diagnosed by immunohistochemistry in SiChuan Provincial People's Hospital from March 2017 to January 2019,and selected 120 cases of female as control group without disease diagnosis evidence during the same period of outpatient physical examination.Oral mucosal epithelial cells and DNA were collected from all these participants to detect the ER?gene Xba?and Pvu?genotyping through real-time fluorescent quantitative PCR technique.Use Thec~2 test to analysis the related risk factors for breast cancer risk and Xba?and Pvu?genotype distribution characteristics.Logistic regression analysis was used to calculate the ratio ratio(OR)and 95%confidence interval(95%CI)to determine the correlation between polymorphism and breast cancer susceptibility,and stratified analysis was conducted according to the menopausal status.According to the immunohistochemical results,the case group was divided into Tri-negative,Her-2,Luminal A and Luminal B.The four molecular subtypes above were stratified by logistic regression analysis and correspondence analysis.Results:After controlling the confounding factors affecting this experiment,based on the analysis of case group and control group,there was no statistical difference between the distribution of three genotypes in ER?gene Xba?loci(wild homozygous type(GG),hybrid type(AG),mutation homozygous type(AA))and the risk of breast cancer(P>0.05)in the total population and groups in various postmenopausal state;ER?gene Pvu?locus mutation of homozygous type(TT)and heterozygous type(CT)with wild homozygous type(CC)significantly increased the risk of breast cancer,OR a value of 7.47(95%CI:3.19~17.59),and according to the hierarchical analysis by states of menopause,the difference is more apparent in premenopausal group,and OR values were 10.99(95%CI:3.13~38.57),4.59(95%CI:1.38~15.28);The conjoint analysis illustrated that individuals with both Xba?homozygous mutations(AA)and Pvu?mutations homozygous type(TT)AATT genotype in total population were prone to suffer from breast cancer,compared to those with other genotypes(P<0.05).According to the stratified analysis of different molecular subtypes of breast cancer in the case group,Xba?loci G allele carriers(wild homozygous type(GG)and heterozygous type(AG))type are at higher risk of Tri-negative breast cancer.The figures for wild homozygous type(GG)and heterozygous type(AG)were the nine times(95%CI:1.07~76.02)and 6.94(95%CI:1.42~34.05)as high as that of the mutation homozygous type(AA);Combined analysis of polymorphisms of the two gene loci found that the risk of Luminal A breast cancer with AATT genotype was higher than that with other genotypes and was not related to menopause(P<0.05).Conclusion:1.There were some correlations between Estrogen receptor alpha gene Xba?and Pvu?polymorphism with breast cancer susceptibility Among women.2.In patients with breast cancer,there were some correlations between Xba?and Pvu?polymorphism with the susceptibility of Luminal A and Tri-negative subtypes.3.The detection of Xba?and Pvu?polymorphism may provide important basis for the preventions and screening and clinical treatment of the breast cancer.
