Objective:This experiment was designed to observe the effect of 1,25-?OH?2D3 on the expression of renal micro RNA-21 in rats and preliminarily explore whether 1,25-?OH?2D3could delay the progression of renal fibrosis by regulating the expression of microRNA-21.Methods:A total of 36 healthy male SD rats were randomly split into three groups:sham operation group?Sham group?,UUO model group?UUO group?,and UUO model treated with 1,25-?OH?2D3 group?treatment group?,with 12 rats in each group.UUO group and treatment group was established by ligating the left ureter.Without ligation,the left ureters were merely dissociated from the rats in the Sham group.After operation,those in the treatment group were given 1,25-?OH?2D3?dissolved in 2ml peanut oil?at a dose of0.06?g/?kg·d?by gavage for 28 consecutive days.Both the Sham group and the UUO group were given an equal volume of peanut oil by gastric perfusion.Six rats in each group were randomly selected to detecte the levels of serum creatinine?Scr?,urea nitrogen?BUN?,calcium?Ca?,phosphorus?P?at the end of day 14 and 28.Meanwhile,the same parts of the left renal tissues were collected to make H-E and Masson staining to observe the kidney pathological changes involving renal tubulointerstitial injury and fibrosis with optical microscope.The real-time PCR was used to measure the expression of microRNA-21 in renal tissues of each group.Results:?1?In the same time period,the level of serum creatinine?Scr?and urea nitrogen?BUN?in treatment group and UUO group increased significantly compared with the sham group?P<0.05?,and the serum creatinine?Scr?level and urea nitrogen?BUN?level was significantly lower in the treatment group than in the UUO group?P<0.05?.With time passing,the level of serum creatinine?Scr?and urea nitrogen?BUN?were no significant difference in the sham group.The level of serum creatinine?Scr?increased gradually in treatment group and UUO group over time,the results were significant difference?P<0.05?.The levels of calcium and phosphorus was not significantly different among the three groups.?2?In the same time period,the degree of renal interstitial fibrosis of UUO group was significantly higher than that in sham group and treatment group.The degree of renal interstitial fibrosis of treatment group was significantly higher than that in sham group.In the same groups,there was no obvious pathological changes in kidney in sham group over time.The degree of renal interstitial fibrosis gradually rising in treatment group and UUO group with the progress of time.?3?In the same time period,the UUO group and the treatment group had relatively higher expression of microRNA-21 in renal tissue than the Sham group,while the expression of microRNA-21 in the treatment group lower than the UUO group,the difference was statistically significant?P<0.05?.With time passing,the expression of microRNA-21 were no significant difference in the sham group and the treatment group.The expression of micro RNA-21 in the UUO group increased over time,the results were significant difference?P<0.05?.Conclusions:?1?1,25-?OH?2D3 probably has the effect of down-regulating the expression of microRNA-21,which inhibits the progress of renal fibrosis in UUO rats.?2?1,25-?OH?2D3 can delay the progress of tubulointerstitial fibrosis in UUO rats.1,25-?OH?2D3 has a protective effect on renal function to a certain extent.?3?The expression of microRNA-21 increased in renal fibrosis in Rats.
