Objective:To investigate the differences and characteristics of braf-activated long non-coding RNA(BANCR)expression in thyroid papillary carcinoma and normal cells.To analyze the role and molecular mechanism of BANCR regulating epithelial mesenchymal transformation(EMT)in promoting invasion and metastasis of thyroid papillary carcinoma.Therefore,the effect of BANCR on the biological function of TPC-1in thyroid papillary carcinoma was revealed.Methods: 1.TPC-1 cells were transfected with si RNA to construct low expression and overexpression cell lines of BANCR,and TPC-1 cell lines of thyroid papillary carcinoma were divided into low expression BANCR group,low expression negative BANCR control group,overexpression BANCR group and overexpression negative BANCR control group.2.The invasion and migration ability of TPC-1 cells in thyroid papillary carcinoma was observed by Transwell chamber invasion test and cell scratch test,and apoptosis was detected by flow cytometry.The difference of cell biological behavior between the low expression group and the overexpression group was analyzed.3.Using protein immunoblot experiment(Western Blot)and real-time fluorescent quantitative PCR(qRT-PCR)to detect epithelial mesenchymal transformation between related molecules(MMP 2,MMP-9,Snail,Vimentin,E-cadherin)express,research BANCR change of expression can lead to MMP 2,MMP-9,Snail,Vimentin,E-cadherin protein and its changes of the mRNA,confirming BANCR in protein level and molecular level on the influence of thyroid papillary carcinoma.Results: 1.In Transwell chamber invasion assay and cell scratch assay,the group with low BANCR expression was stronger in invasion and metastasis than the group with overexpression of BANCR.However,flow cytometry was used to detect cell apoptosis,and it was found that apoptosis was slowed down in the group with low BANCR expression.Experiments show that low expression of BANCR can promote the invasion and metastasis of cancer cells,and prevent apoptosis of cancer cells.2.Western Blot analysis showed that the expression levels of Vimentin,Snailand MMP-2 in the BANCR group were higher than those in the overexpression group,while the expression levels of E-cadherin and MMP-9 were lower than those in the overexpression group.The differences were statistically significant.3.The expression levels of MMP-2,Snail,Vimentin and MMP-9 in qrt-pcr were higher in the low expression group of BANCR than in the overexpression group,while the expression levels of E-cadherin were lower in the former group than in the latter group.The above detection results were statistically significant.Experiments show that low expression of BANCR exerts its role by regulating the related molecules of epithelial mesenchymal transformation Conclusion : Low expression of BANCR enhances the invasion and metastasis of papillary thyroid carcinoma and slows its apoptosis,and promote cell invasion and metastasis by regulating epithelial mesenchymal transformation.The results showed that The change of BANCR expression is involved in the occurrence and development of thyroid papillary carcinoma.It provides a research direction for the next step of targeted drug development.It provides a solid theoretical basis for improving the survival rate and prognosis of patients with thyroid papillary carcinoma.
