Objective: Oral squamous cell carcinoma is one of the most common malignant tumors in the world.Surgical resection is still the most frequently chosen treatment for OSCC patients.Although the surgical method is improving continuously,it is difficult to achieve radical surgical resection due to the fine location and complexity of anatomical structure of OSCC,so the significant improvement was not been seen in the survival rate of OSCC patients.Therefore,it is very important to explore new molecular markers for the early diagnosis,treatment and prognosis of OSCC.In recent years,it has been found that a variety of circular RNAs are abnormally expressed in tumor tissues,which has an important influence on tumor occurrence and progression.However,there are few studies on the relationship between circular RNAs and OSCC.The purpose of this study is to screen circular RNAs differentially expressed in OSCC and to explore the expression level and clinical significance in the diagnosis of hsacirc0086414 in OSCC.Methods:Tumor tissues and adjacent healthy tissues were obtained from 3 patients with OSCC for high-throughput sequencing analysis of circRNAs by Gene Denovo Biotechnology company in Guangzhou.According to the high-throughput sequencing results and literature review,we selected a significantly differential expressed circRNA which has not been studied in oral squamous cell carcinoma as a target circRNA.The relative expression level of hsacirc0086414 was detected in 55 pairs of OSCC tissue samples and cell lines?CAL27,SCC25?by qRT-PCR.Additionally,we analyzed the relationship between the expression level and clinicopathological characteristics.Diagnostic value of hsacirc0086414 in OSCC was assessed by the ROC curve.Functional enrichment analysis and bioinformatics analysis were conducted to further understand the potential functions of hsacirc0086414 and the targeting relationship of hsacirc0086414-miRNAs-mRNAs,respectively.We construct the hsacirc0086414 overexpression plasmid,and transfected the plasmid to CAL27 cells,then detected the plasmid transfection efficiency by qRT-PCR.48 hours after the overexpression of hsacirc0086414,the functional experiment was carried out to test the biological characteristic changes of CAL27 cells.Results: Compared with adjacent healthy tissues,407 differentially expressed circRNAs were identified in tumor tissues by high-throughput sequencing.The expression level of hsacirc0086414 in OSCC tissue samples and OSCC cell lines was discovered to be significantly downregulated compared with the adjacent healthy tissues?AHTs?and normal?HaCaT?cells,respectively.Moreover,hsacirc0086414 expression level was significantly related to TNM stage,tumor size and lymph node metastasis.The area under the ROC curve was 0.749.Hsacirc0086414-miRNAsmRNAs network analysis,GO,DO and KEGG analyses all demonstrated that hsacirc0086414 related to cancer progression to a certain extent.Conclusion: We discovered that hsacirc0086414 might be an essential diagnostic biomarker in OSCC and a target for OSCC therapy.
