Objective: Sjogren's syndrome(SS)is a chronic autoimmune inflammatory disease with dry mouth and eyes or the main symptoms of joint pain.Histopathological manifestations were infiltration of salivary gland interstitial lymphocytes,acinar atrophy,and gland duct damage.Traditional Chinese medicine zedoary turmeric belongs to the group of drugs that break blood and remove blood stasis.At present,zedoary turmeric and its monomers are widely used in experimental research on anti-tumor and anti-inflammatory.NOD/Ltj mice are spontaneous Sj?gren's syndrome model mice,and symptoms of reduced salivary function spontaneously occur at about10 weeks of age.Therefore,this experiment uses NOD/ Ltj mice to study the therapeutic effect of zedoary turmeric on SS,and clarify the effect of zedoary turmeric on NOD / The effects of serum IL-6 and submandibular gland tissue CXCL12 in Ltj mice provide a theoretical reference for the clinical treatment and pathogenesis of SS.Materials and methods: Using 10 SPF-grade 8-week-old ICR mice and 50 SS model NOD / Ltj mice,the ICR mice and NOD/Ltj mice were grouped as follows: A.Blank group(ICR mice + deionized water);B.Model group(NOD/Ltj mice + deionized water);C.Western medicine hydroxychloroquine(NOD/Ltj mice + 60 mg / kg)D.Traditional Chinese medicine zedoary turmeric group(NOD/Ltj mice + 13.5g / kg)E.Chinese medicine Curcuma high group(NOD/Ltj mice + 27 g / kg)F.Curcuma oil group(NOD/Ltj mice + intraperitoneal injection + 60 mg / kg),10 animals in each group,adaptive feeding for two weeks,waiting for onset of 10 weeks of age After 6weeks of administration,the mice were sacrificed after the administration.During the experiment,the whole body condition,saliva flow and blood glucose of the mice were recorded.After the mice were collected,the pathological changes in the submandibular gland tissue of the mouse were evaluated by HE staining and analyzed.The serum IL-6 content was detected by enzyme-linked immunosorbent assay and the expression of CXCL12 in the submandibular gland of the mouse was observed by immunohistochemistry.Results: 1.Compared with the mice in the blank group,the model group and the administration group had slightly yellow fur,reduced eating,no significant increase in body weight,and occasionally scratched the lips and scratched the skin,while the traditional Chinese medicine zedoary turmeric and zedoary turmeric oil groups can improve the above symptoms.2.Compared with the blank group,salivary flow of NOD/Ltj mice began to decrease significantly after 10 weeks of age(p <0.01).After 3 weeks of administration,compared with the model group,the zedoary turmeric group(27g/kg),the zedoary turmeric oil injection group(60mg / kg),and the hydroxychloroquine group(60mg / kg)significantly increased saliva flow(p <0.01,p<0.05),compared with the model group,the saliva flow of the low zedoary turmeric group was improved,but there was no statistical difference,and the saliva flow of the high zedoary turmeric group was higher than the other three administration groups(p<0.01,p < 0.05);After 6 weeks of administration,the saliva flow in each group significantly increased compared with that in the model group(p < 0.01,p < 0.05),the saliva flow in the zedoary high group was higher than that in the remaining group(p <0.01),and that in the zedoary low group was higher than that in the model group(p <0.05),but lower than that in the other groups(p < 0.01,p < 0.05).3.Compared with the mice in the blank group,the weight of the submandibular glands in the model group was significantly reduced(P <0.01),and compared with the model group,each of the curcumin group and the hydroxychloroquine group(60 mg /kg)significantly improved Rat submandibular gland index(P <0.01,P <0.05),improving submandibular gland weight,and the effect of improving the submandibular gland index of the zedoary turmeric group was better than that of the remaining group(P <0.01,P <0.05);compared with the blank group,the spleen index in the model group increased significantly(P < 0.01),while compared with the model group,the spleen index in the zedoary administration group and the hydroxychloroquine group(60mg/kg)significantly increased(P < 0.01,P < 0.05)and decreased the spleen weight.4.In the submandibular gland pathology picture,it can be seen that the submandibular glands of the model group had severe lymphocyte infiltration,the intercellular space increased,and the infiltrated cells were mostly distributed around the catheter and blood vessels.The size of acinars varied,and the ducts of the glands were disorderly arranged.After given zedoary turmeric granules(27g/kg),zedoary turmeric oil and hydroxychloroquine,the histopathological scores of the mice were significantly lower than those of the model group(p <0.01,p <0.05),and the zedoary turmeric low group(13.5g/kg)and the model compared with the group,the pathological tissue score also improved,but there was no statistical difference.5.Correlation analysis was performed between the saliva flow and fasting blood glucose at the end of 16 weeks of age in mice,and the correlation coefficient r =-0.06,p = 0.62 > 0.05,indicating that there was no correlation between saliva flow and fasting blood glucose in mice.6.Pancreatic pathological pictures show that the islet structure of the control group is complete without lymphocyte infiltration,while the model group and each administration group can see lymphocyte infiltration in the islets surrounding the lymphocyte infiltration area < 25%,according to the Schimidt pathological rating standard for pancreatic tissue.It belongs to class I pancreatitis,and there is basically no difference in the inflammation state between the pancreas tissue of each administration group and the model group.7.Compared with the mice in the blank group,the expression of CXCL12 in the submandibular gland of NOD/Ltj mice increased,and each administration group significantly inhibited the expression of CXCL12 in the submandibular gland of the mice(P <0.01,P <0.05).There was a statistical difference in the expression of CXCL12 between the administration groups(P<0.01,P<0.05).8.Compared with the mice in the blank group,the serum IL-6 levels in the model group were significantly increased(p <0.01),while the serum IL-6 levels in the zedoary turmeric administration group and the hydroxychloroquine group were significantly lower than those in the model group(p <0.01,p <0.05),and there was astatistically significant difference in IL-6 levels between the groups administered with zedoary turmeric(p <0.01,p <0.05).In conclusion:1.Zedoary rhizoma has a significant improvement on the secretory function and inflammation of the submandibular glands of NOD/Ltj mice.2.Curcuma Zedoaria can reduce serum IL-6 levels and CXCL12 expression in submandibular glands of NOD / Ltj mice...
