Study On Chemical Composition Of Dictamni Cortex

Dictamni Cortex is a dried root bark of the Dictamnus in the family Rutaceae.According to modern pharmacological studies,it mainly contains alkaloids,limonoids,flavones,coumarins and other chemical components.It can be used to treat Skin diseases such as psoriasis and eczema,it can also be used for the treatment of jaundice,rheumatism and chronic liver diseases.In order to explore the structural diversity of the chemical components in Dictamni Cortex,we extracted and isolated the Chinese medicinal materials,and obtained the structural identification of the monomer compounds to explore its chemical composition further.Crush the Dictamni Cortex raw material,use 70%ethanol-water solvent heating reflux extraction method to extract and concentrate the active ingredients to obtain extracts,and use the petroleum ether,Dichloromethane,ethyl acetate,and n-butanol to extract the crude extract.By using different polar solvents and adjusting different solvent ratios to explore the appropriate column chromatography conditions,the petroleum ether layer and the dichloromethane layer were separated separately,and separated into several small fractions.Each fraction is subdivided,and a variety of separation methods such as thin-layer chromatography,silica gel column chromatography,Sephadex LH-20 column chromatography,ODS reverse phase column chromatography,analytical and preparative HPLC are used to systematically analyze the sample components Isolation.Structural identification of the monomercompounds isolated and purified from Dictamni Cortex,using one-dimensional nuclear magnetic resonance ¹H NMR,¹³C NMR,combined with MS to determine the chemical structure of the monomer substance.This research group separated petroleum ether and dichloromethane extractio layers from 70%ethanol extract of Cortex dictamni,a total of 23 compounds were obtained,and 16 of them were identified.Their structures were identified as dictamine?1??skimmianine?2???-fagarine?3??hoplopine?4??dasycarine?5??glycolone?6??7,8-dimethoxymyrtopsine?7??8,9-dimethoxygeibalansine?8??8-methoxyflindersine?9??3-formylindole?10?,fraxinellone?11??radicol?12??kihadanin A?13?,magnolol?14???-sitosterol?15?,diisobutyl phthalate?16?.Among them,compounds 10,14,and 16 were the first compounds isolated from the genus Dictamnus,and compound 12 was the first compound isolated from the Dictamni dasycarpus.
