| Chapter 1:Fe3O4 magnetic nanoparticles (MNPs) were reviewed mainly on its properties, synthesis, characterization, and applications in medicine, biology and environmental governance. Then the background, research contents and innovation points of this paper were introduced briefly.Chapter 2:After synthesis of Fe3O4 MNPs by chemical coprecipitation method, the obtained MNPs were modified by ethyl silicate and trimethylstearylammonium bromide. As a result, as-prepared TSAB-Fe3O4/SiO2 magnetic composite was characterized by transmission electron microscopy, Fourier transform infrared spectrometer and vibrating sample magnetometer.Chapter 3:The high performance liquid chromatography with fluorescence assay method was established and further applied for the detection of adriamycin hydrochloride (ADR). As a result, the calibration curve, Y=13.33X-5.1008, was obtained by plotting peak area (Y) against the corresponding concentration (X). It exhibited good linearity with a correlation coefficient (r) above 0.9984 and a relative standard deviation below 4.27% in the range from 0.5 to 10.0μg/mL. The limit of detection and limit of quantitation were 5.05 and 16.82 ng/mL respectively. In three different concentrations, the recoveries were in the range of 95.2%-107.3% and 98.2%-101.6% for plasma and urine samples respectively. The proposed method can be applied to detection of ADR from human plasma and urine samples due to its simpleness, accuracy, and specificity.Chapter 4:The TSAB-Fe3O4/SiO2 MNPs was used as the adsorbent for the magnetic solid-phase extraction of ADR in human plasma and urine with high performance liquid chromatography-fluorescence detection. The factors influencing the extraction efficiency were examined including the pHvalue, extraction time and elution solvent etc. The experimental results show that good reproducibility and the recoveries were in the range of 76.5%-94.0% and 77.9%-96.0% for plasma and urine samples with a good reproducibility.Chapter 5:On the basis of the study on synthesis and application of Fe3O4 magnetic nano-composites for the determination of adriamycin hydrochloride in human body fluids, the antibiotic cefoperazone and ciprofloxacin lactate were used as a model drug for studying fluorescence spectrum of drugs and human serum albumin. We are looking forward to this research as a beginning to explore further the interaction between antibiotics medicines and complex biological samples.Chapter 6:This part summarized the research works about magnetic solid-phase extraction based on TSAB-Fe3O4/SiO2 composite for detection of adriamycin hydrochloride in human plasma and urine by HPLC-FLD. In addition, the prepared magnetic composite was expected to be could apply in other aspects. |
