Objective:To explore the effects of enriched environment on the behavior and IL-1?,IL-6 of sodium valproate-exposed autism rats,and to elucidate the improvement mechanism of rich environment on abnormal behavior and brain inflammation.Method:Through intraperitoneal injection of sodium valproate into pregnant rats on the 12.5th day of pregnancy,an autism model mouse was established,and a model group,an experimental group,and a control group were established The rats in the experimental group were given rich environmental intervention 21 days after birth.The model group and the control group did not take any treatment,and only under the same conditions.The experiment was conducted for 3 weeks.After the intervention,the behavioral test of rats in each group was conducted through the bead embedding experiment,the open field experiment,the three-box experiment and the water maze experiment;HE staining and Nissl staining were used to observe the nerve damage and neuron survival in the prefrontal region of the brain;Elisa was used to detect the expression of IL-1? and IL-6 in the serum of each group of rats;Western Blot was used to detect the expression of IL-1? and IL-6 in the prefrontal region of the brain of each groupResults:1.Behavioral testing(1)Marble burying test:Compared with the control group,the number of exposed beads in the model group increased significantly,the difference was statistically significant(P<0.05);compared with the model group,the number of unburied beads in the experimental group was significantly reduced after the experiment,the difference was statistically significant(P<0.05)(2)Open field test:Compared with the control group,the time of the model group in the middle area and the total distance of spontaneous activity increased significantly,the difference was statistically significant(P<0.05);compared with the model group,the time of the rats in the experimental group in the middle area and the total distance of spontaneous activity were significantly reduced,and the difference was statistically significant(P<0.05)(3)Three box test1)Social ability:Compared with the control group,the rats in the model group are more inclined to stay in the empty cage area and the middle partition area,the data results are statistically significant(P<0.05);compared with the model group,the rats in the experimental group had a longer duration of Sniff stranger 1 and the difference was statistically significant(P<0.05)2)Social novelty:Compared with the control group,the rats in the model group are more inclined to Sniff stranger 1 and stay in the blank partition,the data results are statistically significant(P<0.05);compared with the model group,the experimental group of Sniff stranger rats 2 took longer,and the difference was statistically significant(P<0.05)(4)Morris water maze1)Location cruising test:Compared with the control group,the time of the escape latency of the model group was significantly longer,and the data results were statistically significant(P<0.05);compared with the model group,the time of the escape latency of the experimental group was reduced,and the difference was statistically significant(P<0.05)2)Space exploration test:Compared with the control group,the number of rats crossing the platform area in the model group was significantly reduced,and the data results were statistically significant(P<0.05);compared with the model group,the number of rats crossing the platform area in the experimental group increased significantly,and the difference was statistically significant(P<0.05).2.HE staining and Nissl stainingIn HE staining,compared with the control group,the model group had obvious damage to the prefrontal region of the brain,the disordered cell arrangement,the cell structure disappeared,and obvious cell edema and inflammatory cell infiltration were seen;compared with the model group,the injury in the prefrontal region of the experimental group was significantly reduced,the infiltration of inflammatory cells and the degeneration and necrosis of nerve cells were significantly reduced,and the cell arrangement and cell contour were significantly restoredIn Nissl staining,compared with the control group,the frontal lobe neurons of the model group were sparsely distributed,a large number of neurons were damaged,the number of Nissl bodies was significantly reduced,and the difference was statistically significant(P<0.05);compared with the model group,the integrity of the frontal neurons and the number of Nissl bodies in the experimental group were significantly restored,and the difference was statistically significant(P<0.05)3.Expression of serum IL-1? and IL-6Compared with the control group,the peripheral blood inflammatory factors IL-6 and IL-1? in the model group were increased,the difference was statistically significant(P<0.05);compared with the model group,the peripheral serum inflammatory factors IL-6 and IL-1? in the experimental group decreased,and the difference was statistically significant(P<0.05)4.Expression of IL-1? and IL-6 in the prefrontal region of the brainCompared with the control group,the expression of IL-6 and IL-1? in the prefrontal region of the brain of the model group were increased,and the difference was statistically significant(P<0.05);compared with the model group,the expression of IL-6 and IL-1? in the prefrontal region of the brain decreased in the experimental group,and the difference was statistically significant(P<0.05)Conclusion:1.Enriched environment can improve the repeated stereotyped behavior,anxious behavior,abnormal social behavior,and abnormal learning and memory ability of autism rats induced by VPA2.Enriched environment can reduce the prefrontal injury of autism rats induced by VPA and promote the recovery of the number of neurons.3.Enriched environment can reduce the expression of proinflammatory cytokines IL-6 and IL-1? in the prefrontal lobe of autistic rats induced by VPA,and regulate inflammation in the brain.
