Objective To investigate the expression and clinical significance of Septin9 and Cyclin D1 in gastric cancer.Methods A total of 60 cases of carcinoma tissues with gastric cancer were collected in the Second Affiliated Hospital of Anhui Medical University from February 2018 to June2019,and adjacent tissues(>5cm from the edge of cancer tissues)0f the 60 cases were selected as the control group.Patients have complete clinical data.Western blot was used to detect the expression of Septin9 and Cyclin D1 protein in two groups,and the relationship between the expression of Septin9 and Cyclin D1 protein and the clinicopathological characteristics of gastric cancer was analyzed.Moreover,the correlation between Septin9 and Cyclin D1 protein expression was analyzed by Pearson correlation.Results 1.The expression of Septin9 protein in gastric cancer tissues was 0.754±0.179,which was higher than that in para-carcinoma tissues(0.101±0.020),and the difference was statistically significant(P<0.05).2.In gastric cancer tissues with different clinical stages,the expression of Septin9 protein increased with clinical stages,and the difference was statistically significant(P<0.01).3.Septin9 protein and clinicopathological characteristics of gastric cancer: Differentiation degree: The expression of Septin9 protein in the high-medium differentiation group was(0.690 ±0.130)lower than that in the low-differentiation group(0.795±0.163),and the difference was statistically significant(P<0.05);Infiltration depth: The expression of Septin9 protein in T1+T2 group was(0.663±0.105)lower than that in T3+T4 group(0.790±0.150),and the difference was statistically significant(P<0.05);Lymph node metastasis: The expression of Septin9 protein in lymph node metastasis group was0.785±0.170,0.705±0.115 in non-lymph node metastasis group.The difference between the two groups was statistically significant(P<0.05).Clinical stage: The expression of Septin9 protein in stage ?+? was(0.650±0.145)lower than stage ?+?(0.810±0.210),the difference was statistically significant(P<0.05).The expression of Septin9 protein was not significantly correlated with other factors such as gender,age,tumor size,and tumor location(P>0.05).4.The expression of Cyclin D1 protein in gastric cancer tissue was 1.217±0.245,which was higher than in para-carcinoma tissues(0.132±0.038),and the difference was statistically significant(P<0.05).Furthermore,the expression of Cyclin D1 protein increased with clinical stages,and the difference was statistically significant(P<0.05).5.Cyclin D1 protein and clinicopathological characteristics of gastric cancer: Differentiation degree: The expression of Cyclin D1 protein in the high-medium differentiation group was(1.130±0.205)lower than that in the low-differentiation group(1.265±0.275),and the difference was statistically significant(P<0.05);Lymph node metastasis: The expression of Cyclin D1 protein in the group with lymph node metastasis was(1.260±0.280)higher than that in the group without lymph node metastasis(1.105±0.201),the difference was statistically significant(P<0.05);Clinical stage: The expression of Cyclin D1 protein in stage ?+? group was1.095±0.180,1.273±0.270 in stage ?+? group.The difference between the two groups was statistically significant(P<0.05).The expression of Cyclin D1 protein was not significantly correlated with other factors such as gender,age,tumor size,tumor location,and infiltration depth(P>0.05).6.Correlation analysis: In gastric cancer tissues,the expression of Septin9 and Cyclin D1 are positively correlated(r=0.782,P<0.05).Conclusion The expressions of Septin9 and Cyclin D1 proteins in gastric cancer tissues was significantly higher than those in para-carcinoma tissues.Both are related to the degree of gastric cancer differentiation,lymph node metastasis and clinical stage,indicating that Septin9 and Cyclin D1 proteins are closely related to the occurrence and development of gastric cancer.Septin9 is positively correlated with the expression of Cyclin D1 in gastric cancer tissues,suggesting that Septin9 may mediate the progression of gastric cancer by regulating Cyclin D1 levels.Septin9 is expected to become a new tumor marker for gastric cancer.Further research and analysis has certain clinical significance for the diagnosis and treatment of gastric cancer.
