Objective:To investigate the expression of interleukin-11(IL-11)in glioma and its effect on glioma migration and invasion.Method:(1)q RT?PCR was performed to determine the expression levels of IL-11 in11 normal brain tissues,24 glioma tissues and human glioma cell lines.(2)Firstly,IL-11 knockdown plasmids were transfected into LN18 and U251 glioma cell lines.,q RT?PCR assay was performed to determine the expression levels of IL-11 after transfected with sh IL-11.LN18 and U251 glioma cells were tansfected with IL-11 knockdown plasmid or treated with recombinant human IL-11,and Transwell assay detected cell migration and invasion.(3)The relationship between long non-coding RNA HOTAIR(Lnc RNA HOTAIR)and IL-11 expression level in glioma was analyzed by TCGA database.To further investigate whether HOTAIR was a functional target of Il-11,q RT?PCR was used to detected the expression of HOTAIR by transfection with IL-11 knockdown plasmid or rh IL-11.(4)To find out if HOTAIR is involved in IL11-mediated the migration and invasion of the glioma cells synergistically,LN18 and U251 cells were co-transfected with sh-IL11 and pc DNA-HOTAIR,and then the migration and invasion ability was verified by transwell assay.Results:(1)The expression of IL-11 mRNA in glioma tissue was higher than normal brain tissue.Similarly,the expression of IL-11 in glioma cell lines was higher than normal astrocytes.(2)Transwell assays showed that IL-11 knockdown inhibited invasion and migration of glioma cells.In addition,the migration and invasion ability was upregulated after treated LN18 and U251 cells with rh IL11(60ng/ml)for 48 hours.(3)There was a significant positive correlation between IL-11 and Lnc RNA HOTAIR in glioma as predicted by TCGA database.q RT-PCR assay result showed that HOTAIR expression was decreased after transfection with sh IL-11 in glioma cells,but increased after transfection with rh IL-11.(4)Transwell assay results revealed that sh IL-11 suppressed the migration and invation of both LN18 and U251 glioma cells while HOTAIR reversed the decrease.Conclusions: In summary,we reported that IL-11 wae highly expressed in glioma tissues and glioma cell lines.Whereas knockdown of IL-11 inhibits glioma cells migration and invasion.Furthermore,IL-11 may regulate the glioma malignant phenotype by Lnc RNA HOTAIR.
