| Long non-coding RNAs(lncRNAs)have emerged as a new and crucial layer of gene regulation in recent years and regulate various biological processes such as carcinogenesis and metastasis.LncRNA HOTAIR,an oncogenic lncRNA,is involved in human tumorigenesis and dysregulated in cervical cancer.Megakaryoblastic leukemia 1(MKL1),as a transcription coactivity factor,involved in cancer metastasis and cell differentiation.However,the precise mechanism of biological roles of HOTAIR and MKL1 in cancer cells remain unclear.In this study,we found that MKL1 plays a role in inducing the migration and invasion of cervical cancer cells,in addition,the expression of MKL1 decreases after knockdown of HOTAIR,and inhibits the migration and invasion of HeLa cells by qPCR,Western blotting,wound healing and transwell invasion assays.Meanwhile,which demonstrated that high levels of HOTAIR are closely related to MKL1 in cervical cancer tissues.More importantly,Luciferase reporter assay was used to determine that MKL1 can activate HOTAIR by binding to CArG-box in HOTAIR promoter.These results demonstrate that MKL1 promotes the migration and invasion of HeLa cells.HOTAIR can regulate the expression of MKL1,and MKL1 activates the transcription of HOTAIR,indicating a positive feedback loop between HOTAIR and MKL1.Taken together,these findings indicate that MKL1 is an important player in the function of HOTAIR in cancer cell migration and invasion. |
