Objective: Studies of SLE(systemic lupus erythematosus,SLE)patients with peripheral blood mononuclear cells in mi R-326 and mi R-330-5 p and its target gene expression level of DNMT1,and its relationship with clinical disease activity of SLE patients,preliminary exploration of mi R-326 and mi R-330-5 p regulate the expression of DNMT1 mechanism in systemic lupus erythematosus(SLE),for the diagnosis of SLE and provide reliable laboratory indexes illness interpretation.Methods: First of all,the selection conforms to the diagnostic standard of 72 cases of patients with Systemic Lupus Erythematosus(SLE),according to Systemic Lupus Erythematosus(SLE)patients with Systemic Lupus Erythematosus(SLE)Activity score(Systemic Lupus Erythematosus diseases,Activity Index,SLEDAI)standards,SLE cases can be divided into high Activity group 21 cases(SLEDAI 15 points or more),group activities in 25 cases(15 > SLEDAI 10 points or higher)and low Activity group of 26 patients(> 10 points SLEDAI 5 minutes(or higher).Meanwhile,40 healthy people were selected as the control group to collect peripheral blood mononuclear cells from the healthy group and extract total RNA.The expression levels of mi R-326,mi R-330-5p and DNMT1 m RNAwere detected by using Quentitative real-time PCR(q PCR)technology,the relationship between mi R-326,mi R-330-5p and the expression levels of DNMT1 m RNA was analyzed,and the receiver operating characteristic curve(ROC curve)was constructed.To analyze the diagnostic value of mi RNA-326 and mi RNA-330-5p in SLE.Result: 1.The expression level of mi R-326 in SLE patients was significantly higher than that in the healthy control group(P<0.05).The high activity group was significantly higher than the medium activity group(P<0.05),the medium activity group was significantly higher than the low activity group(P<0.05),and the low activity group was significantly higher than the healthy control group(P<0.05).2.The expression level of mi R-330-5p in SLE patients was significantly higher than that in the healthy control group(P<0.05),significantly higher in the high-activity group than that in the moderately active group(P<0.05),significantly higher in the moderately active group than that in the moderately active group(P<0.05),and significantly higher in the low-activity group than that in the healthy control group(P<0.05).3.The expression level of DNMT1 m RNA in SLE patients was significantly lower than that in the healthy control group(P<0.05),significantly lower in the high activity group than that in the moderately active group(P<0.05),significantly lower in the moderately active group than that in the moderately active group(P<0.05),and significantly lower in the low-activity group than that in the healthy control group(P<0.05).4.mi R-326 was negatively correlated with the expression level of DNMT1 m RNA(r=-0.4836,P<0.001).mi R-330-5p was negatively correlated with the expression level of DNMT1 m RNA(r=-0.5922,P<0.001).5.The ROC curve area(AUC)of the expression of mir-326 in the diagnosis of SLE was 0.8919(95% confidence interval =0.8087 ~ 0.9750).The ROC curve area(AUC)for the expression of mi R-330-5p in the diagnosis of SLE was 0.9028(95% confidence interval =0.8385 ~ 0.9671).6.The correlation analysis between DNMT1 m RNA and SLEDAI showed a negative correlation(r=-0.7004,P<0.001).The correlation between mi R-326 and SLEDAI showed a positive correlation(r=0.5086,P<0.001).The correlation analysis between mi R-330-5p and SLEDAI showed a positive correlation(r=0.7909,P<0.001).Conclusion: 1.The expressions of mi R-326 and mi R-330-5p are increased in SLE patients,while the expression of DNMT1 m RNA is decreased in SLE patients,suggesting that mi R-326,mi R-330-5p and DNMT1 m RNA are related to the pathogenesis of SLE.2.The expression levels of mi R-326 and mi R-330-5p were negatively correlated with DNMT1 m RNA,suggesting their possible involvement in the molecular mechanism of SLE.3.ROC curve showed that mi R-326 and mi R-330-5p had certain diagnostic value for SLE and could be used as auxiliary indicators for the diagnosis of SLE.4.DNMT1 m RNA was negatively correlated with SLEDAI,while mi R-326 and mi R-330-5p were positively correlated with SLEDAI,suggesting that they can reflect the disease activity of patients with systemic lupus erythematosus to a certain extent.
