The Role Of Apatinib In Cervical Cancer-resistant Cell Lines

Objective:To explore the mechanism of apatinib in cervical cancer resistant cell lines.Methods:1.CCK8 method was used to detect the effect of different concentrations of apatinib on the proliferation in cervical cancer-resistant cell lines HeLa/DDP,and the IC50 was calculated as the half inhibitory concentration;2.CCK8 method to detect the cell proliferation of apatinib,apatinib and cisplatin on cervical cancer cisplatin-resistant cell line HeLa/DDP;3.Detection of apatinib and apatinib by flow cytometry 4.Apoptosis of cisplatin-resistant cervical cancer cell line HeLa/DDP by nicotine combined with cisplatin;The expression of multidrug resistance related protein P-gp.Results:1.The CCK8 experiment showed that apatinib dose-dependently inhibited the proliferation of cervical cancer resistant cell line HeLa/DDP.The IC50 of HeLa/DDP was 14.553 ?M2.CCK8 experiment showed that apatinib,apatinib combined with cisplatin can inhibit HeLa/DDP cell proliferation,and apatinib combined with cisplatin significantly inhibited HeLa/DDP cell proliferation;Both apatinib and apatinib combined with cisplatin can induce apoptosis in HeLa/DDP cells,and apatinib combined with cisplatin can significantly induce apoptosis in HeLa/DDP;3.Western blot results showed that apatinib,apatinib combined with cisplatin could not inhibit the expression of multidrug resistance related protein P-gp in HeLa/DDP cell line.Conclusions:1.Apatinib can inhibit the proliferation of cervical cancer cisplatin-resistant cell line HeLa/DDP,and can induce apoptosis;2.For cervical cancer cisplatin-resistant cell line HeLa/DDP,apatinib combined with cis Platinum inhibitory effect is more significant;3.Apatinib can reverse the resistance of cervical cancer cisplatin-resistant cell line HeLa/DDP,but it can not inhibit the expression of drug resistance-related protein P-gp,may be reversed through other signaling pathways and mechanisms Drug resistance,enhance the accumulation of cisplatin in cells.
