Objective:To investigate the relationship between the expression of CD 163 positive tumor-related macrophages and Sema4D protein and clinicopathological characteristics in colorectal adenocarcinoma.Methods:A retrospective analysis was performed on the clinicopathological data of 61 patients who underwent radical surgery for colorectal cancer in the third affiliated hospital of Kunming Medical University from 2016 to 2019.There were 33 males and 28 females with the median age of 60 years.Immunohistochemistry was used to detect the expression levels of CD 163 and Semaphorins 4D(Sema4D)in 61 cases of cancer and 30 cases of distant cancer(5cm above the edge of the cancer).Results:The expression levels of CD 163+tumor-related macrophages in the interstitium of colorectal adenocarcinoma and distant carcinoma were statistically significant(P<0.001).Sema4D was 100%expressed in colorectal cancer tissues(61/61),but not in distant cancer tissues(0/30),showing a statistically significant difference(P<0.001).Spearman correlation analysis of the expression of CD 163+ tumor-related macrophages and Sema4D protein in colorectal cancer tissues showed a positive correlation(r=0.330,P=0.009).T test was performed to analyze the difference in expression level of CD 163+tumor-related macrophages among the groups with high expression of Sema4D.The difference in expression level of CD 163+tumor-related macrophages among the groups with high expression of Sema4D was statistically significant(P<0.05).The expression of CD 163+tumor-related macrophages was related with the differentiation degree,lymph node metastasis and TNM stage(P<0.05),and was independent of gender,age,tumor location,preoperative serum CEA value and distant metastasis.Sema4D protein expression was independent of gender,age,tumor location,preoperative serum CEA value,presence or absence of distant metastasis,degree of differentiation,depth of invasion,TNM stage,and presence or absence of lymph node metastasis.Conclusion:(1)The increased number of tumor-related macrophage infiltration in colorectal adenocarcinoma may increase the malignancy and invasion and metastasis of the cancer tissue;(2)The expression of Sema4D may not be directly related to the further development of colorectal adenocarcinoma;(3)High expression of Sema4D in colorectal adenocarcinoma tissues can promote recruitment of tumor-related macrophages.Sema4D and tumor-related macrophages may have a synergistic effect in the development of colorectal adenocarcinoma.Sema4D may be expected to be a research entry point targeting tumor-related macrophages.
