Objectives:In this study,TMT labeled quantitative protein technology combined with LC-MS/MS technology was used to construct the serum differential protein expression profile of patients with thyroid related ophthalmopathy and normal people,combined with bioinformatics technology system to screen and explore the differential protein of related diseases,and to explore the serum protein difference between patients with thyroid related ophthalmopathy who have normal and abnormal thyroid function.The pathogenesis of the disease provides the basis of protein levelMethods:Three healthy volunteers were collected as the normal control group,with an average age of 35.5 years(26-45 years);six patients with Tao from June 2019 to December 2019 were collected as the case group,with an average age of 37.6 years(25-50 years),including three patients with normal thyroid function as the Tao Group and three patients with abnormal thyroid function as the Tao Group.The serum of each group was collected,labeled and stored in the environment of-80? for testing;protein quality inspection was carried out,followed by protein reduction alkylation and trypsin enzymolysis,followed by TMT reagent labeling,combined with liquid chromatography tandem technology system for protein identification and quantitative analysis,the difference multiple was more than 1.2 times and P value was less than 0.05 to screen up-regulated eggs.At last,bioinformatics analysis was carried out on the differential proteinResults:In this experiment,3265 proteins were identified,including 3172 with quantitative information.Compared with the normal control group,there were 46 differentially expressed proteins in Tao Group with normal thyroid function,32 of which were up-regulated,14 were down regulated,and 136 were down regulated in Tao Group with abnormal thyroid function,106 of which were up-regulated and 30 were down regulated;There were 110 differentially expressed proteins in Tao Group with normal and abnormal thyroid function,27 of which were up-regulated and 83 down regulated.Conclusions:1.In this study,TMT quantitative proteomics technology combined with liquid chromatography tandem mass spectrometry(LC-MS/MS)was used to study serum proteomics of TAO patients for the first time,and the differentially expressed protein profiles of TAO patients were successfully established;2.Compared with healthy people,serum proteins with significant changes and quantitative information were found in TAO patients;3.This experiment screened and identified the differential proteins of serum in patients with normal and abnormal functions in TAO.
