Objectives:1.Clarify the activation of Q-switched 1064nm Nd:YAG laser on autophagy in normal and photoaged mice.2.Clarify the mechanism of Q-switched 1064nm Nd:YAG laser to improve photoaging through autophagy.Methods:1.In the experiment of the Q-switch 1064nm Nd:YAG laser skin rejuvenation group,4 female balb/c mice were randomly selected,and after removing the hair on the back,the back skin was divided into 4 pieces on average.It is 0J/cm2,1J/cm2,2J/cm2,3 J/cm2,of which 0J/cm2 is the control group.After continuous irradiation for 2 months,the skin of the mice is taken for subsequent detection.2.In the experiment of Q switch 1064nm Nd:YAG laser treatment of photoaging mice,randomly select 4 female balb/c mice,remove the hair on the back,and use the solar simulator(UVA+UVB)to build a small photoaging The mouse model was irradiated with ultraviolet rays 3 times a week for 3 months,and the final dose was UVA:181.44J,UVB:9.72J;Q-switch 1064nm Nd:YAG laser irradiation was performed while the photoaging model was established The back skin is divided into 4 pieces on average,and the laser irradiation doses are 0J/cm2,1J/cm 2,2J/cm 2,and 3J/cm2,of which 0J/cm2 is the photoaging control group,and laser irradiation is performed twice a week.After continuous treatment with UV and laser for 3 months,the skin of the mice was taken for subsequent detection.3.After the mice were sacrificed,the skin was taken and tested separately:?Western blot method was used to detect the expression of LC3B,mTOR,p-mTOR,PI3K,p-PI3K,collagen I,MMP-1,TGF-?1,TGF-? Receptor1;?Perform paraffin-embedded sections for HE staining histological observation.Results:1.In the skin rejuvenation experiment,the Q-switched 1064nm Nd:YAG laser may activate autophagy in normal mouse skin through the mTOR-independent pathway,and the expression of p-PI3K,PI3K,p-mTOR,mTOR Up-regulated;the expression of TGF-?,TGF-? Receptor1,type I collagen was significantly up-regulated,but had little effect on the expression of MMP-1.In the HE staining results,we observed that after 1J/cm2,2J/cm2,and 3J/cm2 laser treatment,the collagen fibers of the dermis of the mouse skin increased,arranged more closely,and the thickness of the dermis increased.2.In the experiment of the laser treating photoaging group,the Q-switched 1064nm Nd:YAG laser may activate autophagy in the skin of photoaging model mice through the mTOR-independent pathway,and the expressions of p-mTOR and mTOR have hardly changed.The expression of the total amount of PI3K did not change significantly after the energy treatment of 1J/cm2 and 2J/cm2.After 3J/cm2 laser energy treatment,the level of the total amount of PI3K decreased,while the level of PI3K phosphorylation was 1J/cm2 and 2J/cm2.,3J/cm2 after energy treatment;TGF-?,TGF-? Receptor1,type I collagen expression was significantly up-regulated;UV significantly increased the expression of MMP-1,after Q switch 1064nm Nd:YAG laser treatment,in 2J/cm2,3J/cm2 energy can significantly reduce the expression of MMP-1,to prevent the degradation of collagen.In the HE staining results,we observed that the arrangement of collagen fibers in the dermis of the photo-aged mice was disordered and the dermis became thinner.After Q-switched 1064nm Nd:YAG laser treatment,the collagen fibers in the dermis of the mouse skin increased and the arrangement was closer.The thickness increases.Conclusions:1.The Q switch 1064nm Nd:YAG laser can activate autophagy in the skin of normal mice and photoaged mice.2.The 1064nm Nd:YAG laser of Q switch activates autophagy and up-regulates the expression of TGF-? and TGF-? Receptor1,which in turn stimulates the production of type I collagen to achieve the purpose of improving photoaging.3.The Q-switched 1064nm Nd:YAG laser down-regulated the expression of MMP-1 in the photoaging treatment group,which basically had no effect on the expression of MMP-1 in the normal group.The laser can improve photoaging by reducing collagen degradation.
