Objective:To explore the characteristics of macrophages infiltration in adipose tissue of obese rats and provide a new basis for the prevention and treatment of coronary heart disease.Methods:First,20 male adult SD rats were randomly divided into 10 control rats and 10 experimental rats.The experimental group was fed a diet high in lipid,while the control group was fed a diet with base foods.Two groups of rats kept for 80 days were sacrificed.Adipose tissue of the Inguinal,epicardium and epididymis of the two groups of rats were extracted,and the epicardium adipose tissue was weighed and recorded.Adipose tissue specimens of the three parts were fixed,defatted,dehydrated,embedded in paraffin,and sectioned to make adipose tissue sections.Finally,adipocyte sections were detected by immunohistochemistry with antibody markers of CD68(M),CD80(M1)and CD 163(M2).Macrophages were observed under the microscope and counted and recorded after redyeing with colorant stain and hematoxylin.Results:In the control group,the weight of rat epicardial adipose tissue was low,which was[(0.30 ± 0.14)g],significantly lower than that of the experimental group[(0.45 ±0.20)g].There were differences in macrophages in adipose tissue of the inguinal,epicardial and epididymal regions between the two groups.There were no significant differences in the total number of macrophages,m1-type macrophages,m2-type macrophages,m1-type macrophages,m2-type macrophages,m1-type macrophages,and m1-type and m2-type macrophages in the control group(P>0.05).In the experimental group,the total number of macrophages,m1-type macrophages,m2-type macrophages,m1-type macrophages,ml-type macrophages,m1-type and m2-type macrophages in the adipose tissue of epididymis and epicardium were significantly higher than that in the peripheral inguinal fat,while the m2-type macrophages were significantly lower than that in the peripheral inguinal fat(P<0.05).The number of macrophages,m1-type macrophages,m2-type macrophages,ml-type macrophages,ml-type and m2-type macrophages in the experimental group were significantly higher than those in the control group,while the ratio of m2-type macrophages in the experimental group was significantly lower than that in the control group(P<0.05).Conclusion:There was no difference in macrophages in the adipose tissue around the epicardium,groin and epididymis of normal rats,and the total number of macrophages in the adipose tissue of the three parts was lower than that of the obese rats.The content of epicardial adipose tissue in obese rats was higher than that in normal rats,and the total value of macrophages in epicardial adipose tissue of obese rats was significantly increased,but the number of m2-type macrophages was relatively reduced.Therefore,the transformation of inflammatory ml-type macrophages into anti-inflammatory m2-type macrophages may be helpful for the prevention and treatment of atherosclerosis in obese people,then cutting down the accidence of coronary heart disease in obese people.
