Glucose metabolism disorders refer to the changes in hormones and enzymes related to glucose metabolism and metabolic organs in the body,resulting in disorders of glucose metabolism;and the lipid deposition or metabolic abnormalities of blood and related metabolic organs caused by self or environmental factors Metabolic disorders.Diabetes,non-alcoholic fatty liver,obesity and other diseases are closely related to glucose and lipid metabolism disorders.With the improvement of living standards,such metabolic diseases are more and more common,but the mechanism of glucose and lipid metabolism disorders is yet to be completed.In this paper,dexamethasone induced a model of glucose and lipid metabolism disorder in rat liver,further revealing the pathogenesis of glucose and lipid metabolism disorder.Male SD rats of 180-220g were randomly divided into 5 groups,namely,the blank group(Ctrl)and dexamethasone(DEX)between 7d,14d,21d,and 28d groups(DEX07,DEX14,DEX21,and DEX28).The dexamethasone action group was given intraperitoneal injection of 1 mg/kg dexamethasone.After different administration times,the rats were sacrificed to collect serum,liver tissue and stool samples for subsequent experiments.Detect the content of ALT,AST,ALP,Glucose,FINS,TG,CHOL,LDL and HDL in serum respectively;HE staining to observe liver tissue lesions and fat deposition;16srRNA high-throughput sequencing analysis of intestinal bacteria in rat feces Changes in populations;transcriptome sequencing method was used to analyze the expression of differential genes in rat liver,and related genes were functionally enriched.The results showed that,after DEX,compared with the Ctrl group,the liver index of rats with glucose and lipid metabolism disorders increased significantly;the serum levels of ALT,AST,ALP,Glucose,FINS,TG,CHOL,LDL,and HDL increased high.After 16srRNA high-throughput sequencing,it was found that after the DEX effect,the number of OTUs uniquely obtained in the intestinal flora of rats increased,and the number of microbial species increased;the components of intestinal flora affected the phylum and genus-level related flora.Many flora are positively and negatively related to environmental factors;and have an impact on the Alpha diversity and Beta diversity of intestinal flora.After DEX action,transcriptome sequencing found that the number of differential genes increased as the time of DEX action increased;after the enrichment of GO functions of differential genes,it was found that the differential genes were mainly distributed in molecular function,biological processes and cells to form three GO term Differential genes are mainly enriched in signaling pathways such as cholesterol metabolic pathway,arachidonic acid metabolic pathway and adolescent diabetes pathway.The results prove that dexamethasone induces hepatic glucose and lipid metabolism disorders in rats by affecting liver pathology,glucose and lipid index,intestinal flora and transcriptome level gene changes.
