Objective:Taking the patients with lower extremity atherosclerosis as the research object,the relationship between lower extremity atherosclerosis and apolipoprotein E gene polymorphism was explored by measuring apolipoprotein E genotype in serum of patients and normal people,to investigate the pathogenesis of lower extremity atherosclerosis.Methods:From January 2019 to December 2019,50 patients in the Department of Cardiology and geriatrics of the Affiliated Hospital of North Sichuan Medical College were selected as the disease group.The color Doppler ultrasound of blood vessels in both legs was in line with the diagnosis of atherosclerosis in the lower limbs.Meanwhile,30 healthy people were randomly selected as the control group,all from the health examination center of the Affiliated Hospital of North Sichuan Medical College.The age and gender of the disease group matched with it,while their color doppler of blood vessels was normal.We collected the fasting blood routine in the morning of disease group and control group,extracted the genomic DNA,and analyzed the polymorphism of ApoE gene by real time fluorescence quantification PCR.And collect the biochemical data of patients.At the same time,according to the different phenotypes of ApoE gene,the patients and the control group were divided into three groups:type 2(E2E2+E2E3)?type 3(E3E3)?type 4(E3E4+E4E4).All the data were statistically analyzed with SPSS 22.0,and the design significance level P was 0.05.Results:1.There was no significant difference in age and gender between the case group and the control group(P>0.05),which was comparable;2.In the case group,the ApoE genotype was the highest in E3E3,followed by E3E4 and E2E3,and E4E4 was the lowest.The gene frequencies of each ApoE allele were ?2 9%,?3 75%,?4 respectively 16%,?2<?4<?3;in the control group,the ApoE genotype had the highest frequency of E3E3 and the lowest frequency of E2E2.The gene frequencies of each ApoE allele were?2 10%,?3 86.67%,?4 3.33%,?4<?2<?3.The frequency of ApoE ?4 allele and the proportion of ApoE genotype 4(E3E4,E4E4)in the case group were significantly higher than that in the control group,with statistical difference(P<0.05);3.The levels of TC and LDL-C in patients with ApoE genotype 4 were higher than those in patients with ApoE genotype 2 and 3(P<0.05).Conclusions:1.Apolipoprotein E was polymorphic in patients with lower extremity atherosclerosis,with the highest frequency of E3E3,followed by E3E4 and E2E3,and E4E4 was the lowest.2.The frequency of ApoE ?4 allele and apoE4 phenotype(E3E4,E4E4)in patients with lower extremity atherosclerosis were significantly higher than those in normal people.3.In patients with lower extremity atherosclerosis,the TC and LDL-C levels of apoE genotype 4 were higher than those of type 2 and 3.
