Esophageal cancer is the seventh most common cancer with a high mortality rate.Although the early prevention and treatment of esophageal cancer has improved,its prognosis is still poor.To explore the molecular mechanism of its effects on proliferation,migration,invasion and apoptosis is helpful to reduce the mortality of esophageal cancer.miRNAs are a class of non-coding RNAs with a length of about 21-23 bp,which can specifically identify target gene mRNAs.' The target sites are generally located in the 3'-non-translation region(3'-UTR).By forming double-stranded RNA after recognition with specific mRNAs,miRNAs promote mRNA degradation and inhibit post-transcriptional translation.Current studies have found that miR-149-5p plays an important role in the occurrence and development of a variety of cancers,but the role of miR-149-5p in esophageal cancer is rarely studied,and its molecular mechanism remains to be further discussed.Our project mainly explored the effect of miR-149-5p on the occurrence and development of esophageal cancer and its mechanism through in vitro cell experiments.In order to explore the effect of miR-149-5p on the phenotype of esophageal cancer cells,we overexpressed miR-149-5p in the esophageal cancer cell lines KYSE-150,EC 109 and TE-1,and detected its effects on cell proliferation,migration,cycle and apoptosis.MTT assay and RTCA assay showed that miR-149-5p could inhibit the proliferation of esophageal cancer cells.FACS showed that miR-149-5p induced cycle arrest in esophageal cancer cells.In further molecular experiments,we investigated the effect of miR-149-5p on the protein and gene levels of esophageal cancer cells.Western Blot experiments showed that miR-149-5p could activate AMPK signaling pathway and simultaneously inhibit STAT3 signaling pathway.qRT-PCR results showed that miR-149-5p could induce the expression of AMPK upstream genes and inhibit the expression of various downstream STAT3 genes.In addition,miR-149-5p can inhibit the transcriptional activity of STAT3.In summary,in esophageal cancer cells,miR-149-5p,as a regulatory factor,can inhibit the proliferation and migration of esophageal cancer cells by regulating AMPK/STAT3 signaling pathway,thus exerting the inhibitory effect on esophageal cancer.In the future detection and treatment of esophageal cancer,miR-149-5p may become a new target for the treatment of esophageal cancer,which expands the new thinking of targeted treatment of esophageal cancer.
