Objective:Retrospective analysis and prospective study of SARI cases in Luohe City,Henan Province from 201 7 to 2019.Through the detection,identification and analysis of HRV in the specimens,the prevalence and clinical characteristics of HRV in SARI cases in Luohe City,Henan Province were determined.The distribution of serotypes and the genome-wide characteristics of dominant serotypes accumulate basic data for related research on HRV in Henan Province,and provide a scientific basis for the diagnosis and prevention of HRV.Method:Pharyngeal swab specimens from hospitalized SARI cases in Luohe Central Hospital of Henan Province from October 2017 to June 2019 were collected.Real-time RT-PCR was used to screen the throat swab specimens for HRV,and then HRV-positive specimens were used to amplify the target gene sequence of the VP1 region by nested PCR,and the amplified products were sequenced.The obtained sequence was used to construct a genetic relationship tree with the HRV serotype prototype strains downloaded from Genbank,and representative strains of dominant serotypes were selected for whole genome sequence determination.Software such as Sequencher 5.4.5,MEGA 5.05,and BioEdit 7.0.5 were used for sequence splicing,alignment,and affinity analysis.Results:(1)A total of 73 HRV-positive specimens were detected in 2017-2019 inpatient SARI cases in Luohe City,Henan Province,with a detection rate of 7.19%,the sixth highest rate among respiratory viruses(2)65 VP1 region target gene sequences were successfully obtained from HRV positive specimens,containing a total of 41 different serotypes,two of which are proposed to be divided into new serotypes Bpat107 and Cpat58.The analysis found that the serotype with the highest detection rate in HRV-A was A104;the serotype with the highest detection rate in HRV-C was C2(3)The detection of HRV in Henan Province peaked in summer and autumn,with a clear seasonal distribution trend;the prevalence of HRV-C was delayed compared with HRV-A,and the peak was lower than HRV-A.(4)The prevalence patterns of HRV serotypes in Henan Province are different.Some serotypes such as A104 are detected in a short time;some serotypes such as A82 are detected in consecutive years;some serotypes such as A80 are in intervals Checked out.(5)Studies have shown that most HRV serotypes have been in an epidemic state globally,but a small number of serotypes have gradually ceased to be prevalent;and the distribution of global HRV serotypes has regional limitations.(6)Analysis of HRV-A 104 based on the VP4/2 region,this study divided HRV-A104 into three ABC genotypes.The sequences obtained in this study constituted LH-lineage in the C genotype.There are differences in the time and geographical distribution of different types of HRV-A104.(7)Amino acid site mutation analysis was performed on the sequences of VP4/2 and VP1 regions of HRV-A104,and it was found that there were 5 amino acid positions in the VP4/2 region that were mutated compared to the prototype strain JN562727/USA/2009,and all B genes Compared with the prototype strain,the amino acid at position 106 has changed from V to 1;22 amino acid positions have been mutated in the VP1 region.The sequence in this study is located at LH-lineage,and all D’s have been changed to E’s at the amino acid position 91 compared to the prototype strain.Conclusions:(1)HRV is an important viral pathogen in severe acute respiratory infections,and the detection rate in this batch of samples is sixth.(2)HRV has many serotypes.In this study,a total of 41 different serotypes were identified,two of which were suggested to be divided into new serotypes;suggesting that we have a large HRV variability and we should strengthen the monitoring of HRV.(3)In HRV,HRV-A and C are predominant.The predominant serotype of HRV-A is A104 and the predominant serotype of HRV-C is C2.(4)The study found that HRV-A104 had a small epidemic in Henan Province from 2017 to 2019.HRV-A104 was divided into three ABC genotypes.The sequences in this study formed a LH-lineage on the evolutionary tree C genotype,suggesting that HRV-A104 in this study has evolved compared to other sequences.(5)HRV exists widely worldwide,but its serotypes differ in time and geographical distribution.Figure[twenty five]table[forty one]reference[sixty-three]...
