Objectives: Chrysanthemum is the dried capitulum of Compositae Plants Chrysanthemum morifolium Ramat,which has the functions of liver-pacifying and vision-improving,heat-clearing and detoxicating,and relieving wind and depression.Chrysanthemum has both edible value and medical value.The "Shen Nong's Materia Medica" of Han Dynasty classified Chrysanthemum as the top grade,people believed that long-term use of Chrysanthemum could improve vitality and prolong life.Modern studies have found that chrysanthemum contains many active ingredients such as flavonoids and triterpenes,and has multiple pharmacological effects such as anti-oxidation,anti-inflammatory and liver protection.Chrysanthemum cultivated in Kai Feng has a long history.There are a lot of researches on its ornamental and variety breeding.However,there are few reports about the chemical constituents and medicinal value of Kai Feng Chrysanthemum.This study established D-galactose-induced aging model to explore the anti-aging effects of the alcohol extracts and flavonoids from Kaifeng chrysanthemum on aging model mice,in order to provide some help for the development of new resources of Kaifeng chrysanthemum.Methods and results: Taking Kaifeng Chrysanthemum KFC-6 as the research object,the alcohol extract and the effective part of flavonoids were extracted and prepared.Ultraviolet spectrophotometry was used to determine the content of total flavonoids in the alcohol extract and the effective fraction of flavonoids.The results showed that the total flavonoid content of the alcohol extract was 13.49%,and the total flavonoid content of the effective flavonoid fraction was 52.20%.120 KM mice were randomly divided into 10 groups: normal control group,D-galactose model group,the low,medium and high dose group of KFC-6 alcohol extract(ie CL,CM,CH group,the administered doses were 200 mg/kg,400 mg/kg,800 mg/kg),the low,medium and high dose group of KFC-6 effective fraction of flavonoids(ie HL,HM,HH groups,the doses were 50 mg/kg,100 mg/kg,200 mg/kg),apigenin group(the dose was 50 mg/kg),positive vitamin E group(ie VE group,the dose was 200 mg/kg).The subcutaneous injection of D-galactose method was used to construct aging model,and the normal control group was injected with the same amount of normal saline.At the same time,the model was given by intragastric administration for 6 weeks.Observe and record the changes in the signs of the mice during the administration;Morris water maze test was performed 5 days before the last administration;the organ indexes of the heart,kidney,liver,spleen,and thymus of the mice in each group were measured after the last administration.Morphological changes of rat liver tissue.The results show that the alcohol extract of KFC-6 and the effective part of flavonoids can shorten the escape latency of mice,improve the memory retention ability of the original platform position,improve the decline of learning and memory ability caused by aging,and delay the brain aging of model mice;and It was found that the alcohol extract of KFC-6 and the effective part of flavonoids can improve the signs of aging in model mice;it was further found that the alcohol extract of KFC-6 and the effective part of flavonoids in aging mice can improve the organs of the kidney,liver,spleen and thymus of model mice.The organ index can reverse the decline in immune and metabolic functions caused by aging,and can improve the morphology of liver cells,alleviate liver tissue damage,and thus play a role in delaying organ aging.The above results prove that the alcohol extract of KFC-6 and the effective parts of flavonoids have certain anti-aging effects.Further study and determine the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and the content of malondialdehyde(MDA)in serum,brain and liver tissues of each group of mice;determine each group The levels of inflammatory factors interleukin 6(IL-6)and tumor necrosis factor-?(TNF-?)in serum of mice;Western blotting was used to examine the expression of Nrf2,HO-1,and NQO1 proteins in liver tissues of aging mice Level changes.The results show that KFC-6 alcohol extract and effective parts of flavonoids can increase the activity of endogenous antioxidant enzymes SOD and GSH-Px in the blood,brain and liver of aging mice,reduce the production of lipid peroxidation product MDA,and reduce the inflammatory factor IL-6.The level of TNF-? can effectively reduce the oxidative stress levels in the tissues and serum of D-galactose-induced aging mice,and can inhibit the chronic inflammatory state of aging.The internal mechanism of the anti-aging effect of KFC-6 was studied,and it was found that the alcohol extract of KFC-6 can promote the transfer of Nrf2 to the nucleus,thereby increasing the expression of the downstream targeting genes of Nrf2,HO-1 and NQO1.Conclusions: This study shows that the effective components of KFC-6 alcohol extract and flavonoids improve the signs of aging mice by protecting the body's antioxidant capacity,protect the organs from free radical damage,and then play an anti-aging effect.The anti-aging effect may be related to the activation of the Nrf2/ARE signaling pathway.
