| Objective:Chrysanthemum is the dried capitulum of Compositae Plants Chrysanthemum morifolium.Ramat Chrysanthemum has the function of heat-clearing and detoxicating,wind-heat disperseing,liver-pacifying and vision-improving,which is commonly used to treat sore abscess,deep-rooted boil,lung abscess,and discomfort in the throat,influenza and so on.Chrysanthemum cultivated in Kai Feng has a long history.It has been well-known throughout the country.There are a lot of researches on its ornamental and variety breeding.Local people also have the habit of eating it;the ecological value and the health care value of it have gradually attracted people’s attention.However,there are few reports about the chemical constituents and medicinal value of Kai Feng chresanthemum.In this thesis,reaserching on Bian liang zi yu,a kind of chrysanthemum cultivated in Kai Feng,The flavonoids were enriched,and determineted content,extracted,and investigate the protective effect of flavonoids extracts on SD rats with alcoholic and liver acute injury by carbon tetrachloride.Methods:To determine the content of flavonoids of Chrysanthemum bian liang zi yu ultraviolet by ultraviolet spectrophotometry;investigate the influence factors of flavonoids yield ingredients(alcohol concentration,extraction time,extraction temperature,material liquid ratio)through single factor experiment.using the response surface experiment designed method to the optimized process parameters,and enrich flavonoids ingredients by polyamide.We get rats model with acute liver injury caused by alcohol and carbon tetrachloride,and they were randomly divided into groups:blank control group,model group,the high,medium and low dose group,with alcohol extraction to the dose of 440 mg/kg respectively,220 mg/kg and 110 mg/kg.Yellow ketone group effective parts group(to the dose of 220 mg/kg),positive-controlled protect liver capsule(to the dose of 108 mg/kg),positive protect liver pills(to the dose of 189 mg/kg),eight groups in total.Detection of serum ALT,AST,T-SOD,MDA,pathological HE staining ofliver,observe the pathological changes of the liver.Results and conclusion:The best extraction process of extraction alcohol as Chrysanthemum cultivated in Kai Feng bian liang zi yu:70%ethanol,1:40,extracting 2h 80℃;chrysanthemum flavones content of 12.26%,30.72%ethanol extraction of flavonoids in the flavonoid content,the effective part of flavonoids is 69.13%;Chrysanthemum cultivated in Kai Feng bian liang zi yu alcohol extract and the effective parts of flavonoids were significantly increased the T-SOD of serum in acute liver injury rats due to carbon tetrachloride and alcohol-induced,reduce the ALT,AST(P<0.01)in serum,inhibit the generation of lipid peroxidation MDA(P<0.01)in rat serum.Pathological sections show the alcohol extraction of song dynasty purple yu and effective parts of flavonoids obviously improve the liver tissue cell disorder and inflammatory infiltration phenomenon to play a role in liver protection. |
