Systemic lupus erythematosus(SLE),which is an autoimmune disease,occurs in young women,with showing fever,rash,erythema on the outside and multiple organ damage,immune system disorders on the inside.Kidney injury is the most important and common organ injury,and it often develops into lupus nephritis in later stage.In modern medicine,hormones,immunosuppressants and other drugs are generally used for treatment,which has immediate effect,but accompanied by metabolic disorders,hypertension and many other adverse reactions after long-term medication.Once the drug is stopped,the illness will be repeated or even worse.In traditional Chinese medicine,SLE is characterized by heat,toxin and blood stasis.And it is a disease with deficiency and excess of spleen and kidney.The treatment is usually combined with traditional Chinese medicine,such as detoxification,spot removing,blood activating and blood cooling,and spleen and kidney tonifying.It has both the incidental and fundamental,and has little side effect.Shengma Gegen decoction,which comes from Taiping Huimin Heji Bureau prescription,has the effect of refreshing,relieving muscles,penetrating rash and detoxifying.It is mainly used to treat rash,fever and headache.This treatment principle is consistent with the TCM treatment of SLE.The purpose of this study is to observe the preventive and therapeutic effects of Shengma Gegen decoction on SLE in mice by establishing two kinds of SLE models.Firstly,two kinds of SLE models were established by using pristane and active lymphocyte chromatin.The physiological signs,urine protein datas,serum autoantibody and lg G expression levels,blood cell number changes,blood coagulation state and hemorheology of mice were recorded during the experiment.The pathological changes of kidney and related markers of renal inflammation and fibrosis were detected.To evaluate the preventive and therapeutic effects of Shengma Gegen decoction on SLE by comparing the two methods in mice.Then,the way of action of Shengma Gegen decoction in the prevention and treatment on SLE was explored.The mouse spleen cells were extracted to detect the effect of Shengma Gegen decoction on the phenotypes of T-lymphocytes and B-lymphocytes,level of ROS and the expressions of autophagy related proteins in spleen and kidney.To predict and explain the molecular mechanism of Shengma Gegen decoction inthe prevention and treatment of SLE,network pharmacology analysis was used.The predicted pathways was preliminarily verified by experiments.The experimental results showed that the SLE model induced by pristane was accompanied by skin erythema damage,urinary protein,autoantibody and lg G in serum,and hypercoagulable state.Platelet increased,clotting time was shorted,nephritis and spleen immune were damaged.Inflammation and fibrosis proteins NLRP3,collagen-1 and ?-SMA were increased significantly,meanwhile the expression of CD4,CD8 and CD19 was increased slightly.The SLE model induced by active lymphocyte chromatin was accompanied by the increase of serum autoantibody,lg G and urine protein,appearing hypercoagulable state,and the kidney was slightly damaged,the spleen was seriously enlarged,and the immune related factors of IFN-?,CD4,CD19 and CD20 were significantly increased.Shengma Gegen decoction could effectively prevent skin damage,reduce urine protein and the expression of autoantibodies and lg G in serum,and prevent hypercoagulability.In addition,Shengma Gegen decoction could improve the pathological changes of kidney injury and spleen enlargement in mice,and the related factors NLRP3,collagen-I and ?-SMA were significantly decreased,and the levels of CD19,CD20,IFN-? were decreased.According to the analysis of network pharmacology,the biological process of Shengma Gegen decoction in the prevention and treatment on SLE is mainly related to "signal transduction","inflammatory response" "Response to estradiol","response of cells to hypoxia","response of cells to lipopolysaccharide" and "negative regulation of apoptosis process";the action pathway mainly involves "toll like receptor signaling pathway","PI3K Akt signaling pathway","HIF-1 signaling pathway" and "NF-?B signaling pathway".The experiment results showed that the autophagy functions of spleen and kidney were enhanced,the expression level of ROS was increased,and the expressions of T-lymphocyte and B-lymphocyte were over increased in SLE model mice;the proliferation rate of T-lymphocyte and B-lymphocyte was decreased in a dose-dependent manner,and the effect on B-lymphocyte was more significan;the expression of HIF-1 ?,HO-1,TLR4,PI3 K,Akt,and NF-?B proteins were detected.These results preliminarily verified the role of Shengma Gegen decoction in the prevention and treatment on SLE.To sum up,Shengma Gegen decoction can play a better role in the prevention and treatment on SLE.Regulating the inflammatory environment of the body,alleviating the abnormal blood coagulation,reducing theimmune damage of the kidney and improving the immune imbalance of the body are the main ways to prevent and treat SLE.
