Objectives:1.To explore the efficacy of CIK cells combined with chemotherapy in the treatment of postoperative patients with colorectal cancer.2.To explore the expression of SLAMF7 in CIK cells and colorectal cancer tissues and its relationship with the efficacy of CIK combined with chemotherapy in postoperative patients with colorectal cancer.Methods:1.A retrospective study was adopted,and 137 cases of colorectal cancer radical surgery in the Department of General Surgery,Yan'an Hospital Affiliated to Kunming Medical University,from January 2007 to December 2014 were selected for study.Divided into CIK treatment group(65 cases)and control group(72 cases).The two groups of patients began follow-up after the first postoperative intervention,and the follow-up deadline was October 2018.The t-test and chi-square test were used to compare the clinical data of the two groups.The Kaplan-Meier method was used to draw the survival curves of the two groups according to the group expression.To study the effect of chemotherapy combined with CIK on overall survival(OS)and disease-free survival(DFS).2.CIK cells were cultured,and the expression of SLAMF7 protein in CIK culture was detected by flow cytometry.SLAMF7+CIK cells were sorted by flow cytometry,and the tumor killing efficiency was verified by tumor killing experiment.3.Paraffin specimens of tumor primary sites and adjacent tissues were removed from surgical specimens of patients receiving chemotherapy combined with CIK treatment.Immunohistochemistry(IHC)technology was used to slice the above paraffin specimens and SLAMF7 staining was conducted,and staining results were independently scored and localized by 2 pathologists respectively.Detection OF SLAMF7 in colorectal cancer tumor tissue and tumor adjacent tissue expression,SLAMF7 in tumor tissue positive expression rate,analysis of SLAMF7 expression and correlation of clinicopathological factors.According to the SLAMF7 expression situation using Kaplan-Meier method to draw the survival curve of patients,comparison of survival curve differences by log-rank test,study the relationship between SLAMF7 expression and patients' total survival(OS)and progression-free survival(DFS).Results:1.CIK cells combined with chemotherapy significantly prolonged the survival time of patients with OS and DFS,.The increase of CIK treatment cycle was positively correlated with the better long-term effect of CRC patients,but there was no significant difference.Postoperative CIK cells combined chemotherapy could significantly prolong OS and DFS in patients with II stage CRC.2.During the induction of CIK cells,the proportion of SLAMF7 positive cells was 5.24%on the 0th day,30.44%on the 7th day,and 64.54%on the 14th day The expression of SLAMF7 protein increased gradually with the induction of CIK.After flow cytometry sorting,the tumor killing activity of CIK cells with high expression of SLAMF7 protein was significantly stronger than that of CIK cells with low expression of SLAMF7 protein(P<0.05).3.SLAMF7 protein was expressed in colorectal tumors and paracancerous tissues,and the expression of SLAMF7 in tumor tissues was lower than that in paracancerous tissues(P=0.0114).The expression of SLAMF7 protein in?-? stage colorectal cancer was significantly decreased(P=0.0338).4.The high expression of SLAMF7 protein in tumor tissues of CRC patients who received CIK combined with chemotherapy corresponded to better long-term survival,but there was no statistical difference(P>0.05).Conclusions:1.CIK combined with chemotherapy can significantly prolong the total OS and DFS of patients with II stage CRC.Increasing the treatment cycle of CIK is positively related to the better long-term effect of CRC patients.2.The expression of SLAMF7 protein on the surface of CIK cells increased gradually during the induction in vitro.3.The expression of SLAMF7 in tumor tissues of patients with III-IV CRC was significantly decreased.4.The patients with high expression of SLAMF7 protein in CRC tumor tissue are more effective to receive CIK combined with chemotherapy after operation.
