[Objective]:To detect the methylation of the CpG island region in the angiotensinogen(AGT)gene promoter of the peripheral blood samples of selected non-H-type hypertensive people,H-type hypertension people and healthy people To compare and analyze the differences in methylation levels in this region among the three groups of people,explore the methylation level characteristics of the AGT gene promoter region in patients with H-type hypertension,and provide new ideas for the prevention and treatment of H-type hypertension from the genetic level.[Methods]:Randomly selected 11 outpatients of primary non-H-type hypertension and newly diagnosed H-type hypertension in the outpatient department of Kunming First People's Hospital from February 2018 to February 2019 24 cases,and 13 healthy people in the medical examination center of Kunming First People's Hospital in the same period.Among them,the healthy population was used as the control group,and the H-type hypertension population and the primary non-H-type hypertension population were taken as the research objects.All participants gave informed consent,participated voluntarily,and signed an informed consent form.Collect the general information of the selected subjects(name,gender,age,blood pressure,weight indicators,drinking history,smoking history,diabetes history,etc.);and laboratory tests(blood homocysteine,renal function,blood sugar,blood lipids)Wait).Pyrosequencing was used to detect the methylation of CpG island region in the AGT gene promoter of peripheral blood samples of the selected subjects.The experimental data was analyzed using SPSS21.0 software.The methylation level is expressed as a percentage,and the measurement data is expressed as(X±S,mean ± standard deviation).Firstly,the data were tested for normal distribution and homogeneity of variance.The comparison between multiple groups used single-factor analysis of variance,and the comparison between groups used the least significant difference method(Least Significant Difference,LSD).All analyses set the P value as a two-sided distribution,with ?=0.05 and P<0.05 indicating statistical differences.[result](1)There were no significant differences in the methylation level of each of the 5 CpG sites in the AGT gene promoter region of the 3 groups(P>0.05);(2)5 cases of H-type hypertension The average methylation level of CpG sites was lower than that of healthy controls,and the difference was statistically significant(P<0.05).[Conclusion]:The average methylation level of the AGT gene in the H-type hypertension case group is lower than that of the healthy people.It can be expected to be used as a breakthrough point at the genetic level in the prevention and treatment of H-type hypertension;The synergy of multiple abnormal methylation genes is not yet known,and further exploration is needed.
