Objective:In recent years,many studies have shown that IQGAP3(IQ Motif Containing GTPase-Activating Protein 3)plays a very important role in the progress of several human cancers.The purpose of this study was to investigate the expression and contribution to the regulation of biological function in Prostate cancer.Methods:.We first evaluated the expression level of IQGAP3 in prostate cancer by immunohistochemistry and comparison of relevant information in the TCGA database(The Cancer Genome Atlas Program).Secondly,the expression of IQGAP3 in cell lines was down-regulated by lentiviral transfection technology,and the success of down-regulation was verified by PCR and Western blot.Finally,relevant functional tests(cell proliferation experiments,scratch experiments and flow cytometry)were used to explore the effect of IQGAP3 on prostate cancer cells Effects of biological behaviors on proliferation,migration,and apoptosis.Results:IQGAP3 was significantly up-regulated at the tissue level of prostate cancer,which is consistent with the conclusions obtained from the comparison of the TCGA database.Prostate cancer cell line DU145-IQGAP3-shRNA whose expression was down-regulated by IQGAP3 and the control group DU145-NC were successfully constructed by lentiviral transfection technology.The results of CCK-8 cell proliferation experiment showed that when the cell proliferation test was performed at three time points:24h,48h,and 72h,the proliferation level of the DU145-IQGAP3-shRNA group was significantly lower than that of the DU-145-NC group.The wound healing experiment(scratch test))The results showed that within 72 hours after the scratch,the total relative cell migration distance was significantly shorter in the DU145-IQGAP3-shRNA group than in the control group.Flow cytometry experiment results showed that 48 hours after virus transfection,DU145-IQGAP3-shRNA group induced more significant apoptosis than DU-145-NC group cells.Conclusions:The expression of IQGAP3 in prostate cancer tissues is higher than that in adjacent tissues.Down-regulating the expression of IQGAP3 in prostate cancer cell line DU-145 can significantly inhibit cell proliferation,migration ability and obviously promote apoptosis.
