Background and Objective Prostate cancer is a common malignant tumor in men over 50 years old.It is one of the leading causes of cancer-related death in men in the West.In recent years,the incidence and mortality rate in China have been increasing year by year.Although prostate specific antigen(PSA)detection has greatly improved the early diagnosis of prostate cancer,the five-year survival rate of early prostate cancer has reached nearly 100% 4,but the patient's later progress is more limited to treatment.Castration-resistant prostate cancer(CRPC).Therefore,it is particularly important to study early prevention and advanced treatment of prostate cancer.Because of the low degree of differentiation of tumors that have developed resistance to a variety of cytotoxic drugs,the use of natural compounds for chemoprevention and treatment has received considerable attention.Cyanidin,as an important member of naturally occurring polyphenolic compounds,have strong reducibility.The ortho-diphenol structure on the B ring is the main active structure that inhibits tumor growth and metastasis.The level of reactive oxygen species(ROS)exerts an antioxidant effect5.Studies have reported that cyanidin play a role in promoting apoptosis and inducing differentiation in prostate cancer cells LNCap and DU145.However,the effects and mechanisms of cyanidin on proliferation,invasion and migration of prostate cancer cells need to be further explored.Therefore,in this study,we used the androgen-independent prostate cancer cell line PC-3 as the basis for further study to investigate the effects of cyanidin on proliferation,migration,invasion and apoptosis of PC-3 cells and their possible effects.Mechanisms are important for guiding prostate cancer prevention and treatment.Methods 1.To study the effect of cyanidin on the proliferation of PC-3 cells.The CCK-8 assay was used to examine the effect of cyanidin on PC-3 proliferation.Scratch test and Transwell test were used to detect the migration and invasion ability of PC-3.2.Observe the effect of cyanidin on cell cycle and apoptosis of PC-3.PC-3 cells treated with different concentrations of anthocyanin were cultured for 48 hours,and the apoptosis and cell cycle of PC-3 cells were detected by flow cytometry.3.Detection of changes in ROS levels in PC-3 cells.The detection of ROS levels in PC-3 cells treated with different concentrations of cyanidin by flow cytometry was used to understand the changes in oxidative stress environment in PC-3 cells.4.Detection of changes in the expression level of SEPW1 in PC-3 cells.The changes of selenoprotein W in selevar by qRT-PCR and WB were used to investigate the molecular mechanism of anthocyanin inhibiting PC-3 cell proliferation and promoting apoptosis.Results 1.Cyanidin have obvious inhibitory effects on proliferation of prostate cancer cell line PC-3 in a concentration-dependent manner.2.When the anthocyanin concentration is high,it can promote the apoptosis and block the cell cycle in the G1 phase.3.The intracellular ROS level decreased with the increase of anthocyanin concentration,showing antioxidant effect.4.At the mRNA and protein levels,cyanidin reduced the expression of SEPW1 in PC-3 cells.Conclusion 1.Cyanidin can significantly inhibit the proliferation,migration and invasion of PC-3 cells,promote the apoptosis of PC-3 cells,and block the cell cycle of PC-3 in G1 phase;2.Cyanidin can reduce ROS levels and down-regulate the expression of SEPW1 to inhibit PC-3 cell proliferation,migration and invasion.
