Background:Programmed death factor-1(PD-1)and its ligand PD-L1 could not only promote tumor cells to achieve immune escape by reducing immune response and suppressing T cell activation,but also reduce immune attack to prevent the occurrence of autoimmune diseases.According to previous literature,we had learned that the exact association between PD-1/PD-L1 single nucleotide polymorphism with autoimmune diseases(type 1 diabetes,systemic lupus erythematosus,ankylosing spondylitis,etc.).Inflammatory bowel disease(IBD)as idiopathic inflammatory diseases involving the gastrointestinal tract had not been reported in the literature about their association with the single nucleotide polymorphism of PD-1/PD-L1.Objective:To assess the association between PD-1/PD-L1 single nucleotide polymorphism with IBD susceptibility in Chinese patients.Methods:Eighty-three IBD patients and 83 healthy controls participated in this study.Four ml peripheral blood sample was collected from each subject.Genomic DNA was amplified by polymerase chain reaction(PCR).Pyrosequencing was performed to genotype five selected SNPs of PD-1 and PD-L1.The experimental data were summarized and further stratified into Crohn’s disease(CD)group and ulcerative colitis(UC)group,male patient group and female patient group.Hardy-Weinberg test was used to measure the representativeness of the control group,and the data was statistically analyzed using the SPSS 24.0 chi-square test.P<0.05 was indicated that there are statistical differences.Results:The AA genotype of rs 10204525 was more in the IBD group than in the healthy group(P=0.036).In addition,the AA genotype was more in the Crohn’s disease(CD)group than in the ulcerative colitis(UC)group(P=0.002),and more in the male group than in the female group.Conclusion:The results indicated that the single nucleotide polymorphism of rs 10204525 in PD-1 was significantly associated with IBD susceptibility,especially in the CD patient population and the male patient population. |
