Malignant tumor is now primary disease threatening human health in the world.Bladder cancer is the worldwideworst one of the first ten common cancer.The author detected the CD molecules expression level in primary bladder cancer cells.We preliminarily found that the expression of CD46,CD59,CD70 and CD164 four CD molecules were showing significantly different than othes,which indicated they could be potential bladder cancer stem cell markers.When we identified in bladder cancer cell line,we identified the mRNA expression level of the four CD molecules.The results showed that the mRNA expression level of the four CD molecules in EJ CD44+cells was higher than EJ CD44-cells,in other words,the mRNA expression level of the four CD molecules in EJ bladder cancer stem cells is significantly higher than the EJ bladder cancer non-stem cells.Back to the primary bladder cancer sample we found that the consistency in CD46 and CD59 molucules.We further identified the function in positive cells screened by the four CD molecules and the results confirmed our judgement.According to the results,EJ cells expressing positive CD46 or CD59 showed outstanding ability in proliferation and drug resistance.Summarize the above results,CD46 and CD59 molecules are potential bladder cancer stem cell markers.The author further identified the mRNA expression level in primary bladder cancer samples by making the gene chip to explore the mechanism of the stemness maintenance.2112 high expression genes and 2438 low expression genes were screened.We further enriched 34 genes by KEGG and identified out 24 genes by GEO database.Finally,8 genes were confirmed by qPCR.The results show that the 8 genes could be related to the mechanism of the stemness maintenance.
