Objective: to establish the acne model in rabbit ears by using modern medicine and animal experiment,and to study the protein expression level of gsta1 and the content of substance P in serum in the process of acne,so as to further study and reveal the mechanism of danlianxiaocuosan in the treatment of acne.Methods: the experimental rabbits were randomly divided into six groups: blank group,positive control group,model group,high dose group,middle dose group and low dose group.After the model was made according to the plan of each group,the corresponding intervention treatment was given to observe the pathological changes of the skin in the model of rabbit ear,the expression of gsta1 protein was measured by immunohistochemistry,and the content of substance P in serum was measured by enzyme immunoassay.Results: 1.In appearance,the acne models of each group were significantly improved after administration.2.In gsta1 index,the blank group,model group and other five groups have differences,and there is statistical significance(P(27)0.01);the positive control group and the middle dose group of danlianxiaocuosan have significant differences(P(27)0.05),and there is no significant difference with the high and low dose groups of danlianxiaocuosan(P(27)0.05);the middle dose group of danlianxiaocuosan has significant differences with the high and low dose groups of danlianxiaocuosan The difference(P(27)0.05)showed that the effects of Dan Lian Xiao Cu O San and Ke Ke Yin ketone gel on the expression of GSTA1 protein were all down regulated,and the middle dose group of Dan Lian Xiao Cu O San was better than that of keketone ketone gel(P(27)0.05).3.SP index: there was significant difference between the blank group and the middle dose group of danlianxiaocuo powder(P(27)0.05).Compared with the other groups,the content of SP was significantly different(P < 0.01);there was significant difference between the model group and the other five groups(P < 0.01);there was no significant difference between the positive control group and the high and low dose group of experimental drugs(P(27)0.05),and there was significant difference between the positive control group and the middle dose group of danlianxiaocuo powder(P < 0.01))There was significant difference between the middle dose group and the high and low dose group(P(27)0.05).The results showed that the dosage of Dan Lian Xiao Cu O powder was better than that of keketone ketone in reducing the content of SP.Conclusion: 1.External administration of danlianxiaocuosan can obviously alleviate and improve the skin lesions caused by acne.2.Danlianxiaocuosan can interfere with the expression of gsta1 protein,which proves that reducing gsta1 is a mechanism of interfering with the development of acne.3.Danlianxiaocuosan can reduce the content of SP,thus inhibiting the development of acne inflammation,which proves that reducing the expression of SP is a mechanism of action in the treatment of acne.
