Objective:To investigate the effect and mechanism of serum containing Yinchen Siling granules on the migration ability of Bone Marrow Mesenchymal Stem Cells.Methods:The contents of SDF-1 in peripheral blood and liver tissues were determined by constructing the rat model of acute liver failure.BMSCs were extracted and interfered with at low,medium and high concentrations of the drug-containing serum of Yinchen Siling granules to observe their cell activity,migration ability and CXCR4 protein expression.Results:(1)The 48 h survival rate of rats in the model group was 33.3%.(2)48h after model construction,serum SDF-1 level of ALF rats was lower than that of the normal group(P<0.05),and SDF-1 was significantly expressed in liver(P<0.05),mainly in bile duct epithelium and oval cells,but not in mature liver cells.The concentration difference of SDF-1 was formed between serum and tissue.(3)There was no difference between the low and medium dose groups and the control group(P > 0.05),but there was a difference between the middle and high dose groups(P < 0.05).There was significant difference between the medium dose group and the high dose group(P < 0.05),and the cell activity in the high dose group was higher than that in the medium dose group(P < 0.05).(4)The number of cell migration in the groups with high and medium concentrations of drug-containing serum was higher than that of the groups with low and control concentrations,the group with high concentration was higher than that of the group with medium concentration(all P <0.05).There was no significant difference between the control group and the low-concentration group(P> 0.05).The blocker group was lower than the control group(P<0.05).(5)Compared with the control group,the expression of CXCR4 protein in the drug-containing serum group was increased,the expression of CXCR4 protein in the high-concentration group was the highest,the difference between the groups was statistically significant(P<0.05).The expression of CXCR4 protein in the blocker AMD3100 was significantly reduced,and the difference was statistically significant(P<0.05).Conclusion:In the serum of rats with acute liver failure,the expression of sdf-1 was low in the serum,and the expression of SDF-1 was high in the liver tissues.The difference in the concentration of SDF-1 in peripheral blood and liver tissues was conducive to the homing of BMSCs to the liver.The drug-containing serum of yinchen siling granules can promote the proliferation and migration of BMSCs.The drug-containing serum of yinchen siling granules can improve the migration ability of BMSCs,and the mechanism may be related to the SDF-1 /CXCR4 axis.
