Research background:China is one of the countries with the highest burden of cardiovascular diseases in the world.Currently,atherosclerotic cardiovascular disease(ASCVD)mainly includes ischemic heart disease and ischemic stroke.ASCVD in China mainly presents the following characteristics:continuous rapid growth,large regional differences,the significant increase in the occurrence of senile cardiovascular diseases,increasing incidence of minor cardiovascular diseases,extremely high out-of-hospital mortality of ischemic heart disease,and extremely severe prevalence of hypertension,diabetes and hyperlipidemia.Coronary atherosclerotic heart disease is characterized by high morbidity and mortality,and the age of onset tends to be younger and younger.Therefore,coronary heart disease is a serious threat to human health.Method for the treatment of coronary heart disease in addition to the reasonable exercise,diet,and quit alcohol life outside intervention and strict drug intervention,for complex coronary heart disease(CHD),percutaneous coronary artery stent implantation(PCI)and coronary artery bypass curative effect is obvious,including PCI surgery because of its less trauma,easy to operate,have a significant effect for improving the prognosis of symptom and the characteristics of,so the PCI surgery in the treatment of coronary heart disease(CHD)having the effect that cannot replace.However,the current serious problem is that there is a high proportion of stent restenosis after PCI.Although the proportion of stent restenosis after PCI has decreased with the use of drug stents,it is still no less than 5%-10%.Previous studies have shown that restenosis in the stent is related to vascular remodeling,inflammatory response,intimal hyperplasia,etc.,but there is still no good plan to reduce the incidence of restenosis(the relationship between IL8 and IL18 levels and stent restenosis after PCI).Current studies have found that restenosis after stenting may be closely related to intimal hyperplasia caused by the proliferation of vascular smooth muscle cells.This paper will study the effect of dioscin on vascular smooth muscle proliferation through both cell and animal experiments,hoping to provide some guidance for clinical practice.Research purpose:1.To investigate the inhibitory effect of dioscin on vascular smooth muscle proliferation,and to confirm the preventive effect of dioscin on vascular restenosis.2.To investigate whether dioscin can inhibit the proliferation of rat vascular smooth muscle cells by promoting autophagy.Research methods:The role of dioscin in inhibiting the proliferation of vascular smooth muscle cells was verified by in vivo balloon injury model and in vitro vascular smooth muscle cell proliferation model.Platelet derived growth factor was used to treat vascular smooth muscle cells in rats to construct vascular smooth muscle proliferation models,and the carotid artery balloon was used to construct balloon injury model in SD rats.Research results:According to the EDU and CCK-8 experiments,it was found that diosgenin could inhibit vascular smooth muscle proliferation in a concentration-dependent way,and the effect was the best when it was 4?mol/L(P<0.05).In animal experiments,HE staining in sections showed that diosgenin could effectively inhibit the proliferation of carotid intima after balloon injury(P<0.05),and the proliferative intima tissue could be identified as vascular smooth muscle cells by immunohistochemistry.By Western Blot,protein of proliferation and phenotypic were inhibited in vivo and in vitro(P<0.05),and the protein of autophagic flux of vascular smooth muscle cells stimulated by PDGF was unobstructed(P<0.05).The results showed that the blocked autophagy could promote the proliferation and phenotypic transformation of vascular smooth muscle cells.In the scratch test,diosgenin showed a concentration-dependent inhibition of vascular smooth muscle migration(P<0.05).Moreover,the obstruction of autophagic flux enhanced the migration ability of smooth muscle cellsResearch conclusions:Our results indicate that dioscin has an anti-proliferation effect on PDGF-BB-stimulated VSMC proliferation by inducing autophagy,suggesting that autophagy may play a role in vascular lesions.This study provides new insight into the effect of dioscin on the proliferation of vascular smooth muscle cells and supports the idea that dioscin may play a role in vascular restenosis.
