BACKGROUNDChronic obstructive pulmonary disease(COPD)is a chronic pulmonary disease,which is characterized by incompletely reversible airflow restriction and persistent respiratory symptoms,usually progressive,and it is also a common,expensive and preventable chronic respiratory disease.COPD has a high incidence rate in China,causing a serious social burden and endangering human health.It has a progressive development,even if quit smoking or use the existing treatment measures,the disease will still further worsen,and there is still a lack of effective means of early prevention,diagnosis and treatment of COPD.In order to solve this problem,a large number of studies have tried to find the risk factors that lead to COPD in order to obtain early detection and early prevention.The results show that the pathogenesis of COPD is the result of the interaction between genetic genes and environmental factors,and glutathione S-transferase Pl(GSTPl)is one of the important biotransferase,which is an important member of glutathione S-transferase(GSTs)superfamily.Glutathione s-transferase(GSTs)is a phase II drug metabolic enzyme that catalyzes the binding of reactive intermediates(such as environmental toxins,oxidative stress products)to glutathione(GSH)for detoxification to produce less reactive water-soluble compounds,which plays an important role in protecting cells from the damage of endogenous and exogenous compounds and preventing the occurrence of COPD.Due to the polymorphism,GSTs can change to its activities and lead to increased susceptibility to toxic compounds,which is closely related to the occurrence of chronic obstructive pulmonary disease(COPD).Therefore,on the basis of previous studies,this study further explored the relationship between polymorphism of rsl695 locus in GSTPl gene and COPD susceptibility,and for the first time explored the relationship between polymorphism of rs4147581 locus in GSTPl gene and COPD susceptibility.This study is helpful to explain the role of COPD related genes and environmental factors in the development of disease,and provide early prevention plan for COPD susceptible population,so as to lay a theoretical foundation for early prevention and effectively reducing the incidence rate of COPD.This study focuses on the association between polymorphism of rsl695,rs4147581 locus in glutathione S-transferase P1 gene and susceptibility to chronic obstructive pulmonary disease,so as to provide early warning for the susceptible population of COPD.METHOD(1)A case-control study was conducted,224 COPD patients who were hospitalized in Huaihe Hospital of Henan University from October 2017 to December 2018 were selected as the case group,and 224 healthy adults matched with the sex and age of the case group were collected as the normal control group.(2)Polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP)analysis was used for the rsl695 polymorphism of GSTPl gene,and the distribution difference of genotype frequency between case group and control group was analyzed.(3)Polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP)and sequencing were used to genotype the rs4147581 locus of GSTPl gene,and the distribution difference of its genotype frequency between the case group and the control group was analyzed.RESULT(1)The smoking rate of the case group was higher than that of the control group,which indicated that smoking was the main risk cause of the disease.(2)The genotype of GSTPl at rs4147581 site was associated with susceptibility to COPD,and the genotype with G allele can increase the risk of COPD,while the polymorphism of rs1695 locus of GSTPl gene has no correlation with the occurrence of COPD.(3)GSTPl gene at rs1695 site and rs4147581 site smokers who carried G allele and C allele were at increased risk of COPD by stratifying smoking.CONCLUSIONThe polymorphism of rs4147581 locus of GSTPl gene is associated with the risk of COPD,while the polymorphism of rsl695 locus of GSTPl gene has no correlation with the occurrence of COPD.
