Aims:To detect the difference of genomic DNA N6-methyladenine between patients with essential hypertension and healthy volunteer,and to preliminarily explore the related pathogenesis.Methods:We collected the peripheral blood from primary hypertension tertiary patients(experimental group)and healthy volunteers(control group)in Shen Zhen;DNA was obtained by centrifugation and qualified DNA samples were extracted to construct library.CASAVA software was used to obtain base information and search for methylation sites.MACS software was used to model the methylation sites,extract the related peaks of methylation difference sites,which was used to obtain the information of methylation difference sites.The methylation difference sites were analyzed by statistical software,and the qualified results were annotated by bioinformatics software.Results:1.Methylation sites: The identified methylation sites in patients with primary hypertension group(31,436,312)were higher than those in the healthy control group(29,235,794),and the proportion of methylation sites in the reference genome,the number of gene repeats and the distribution in the genome in the peripheral blood of patients with primary tertiary hypertension were higher than those in the healthy control group(P < 0.05).2.Methylation difference sites: There were 367,117 methylation difference sites in patients with primary hypertension,and 37,564 methylation difference sites in gene functional areas.There were 235,336 different methylation sites in the healthy control group,and 32486 different methylation sites in the gene functional area.The total number of methylation difference sites and the number of functional difference sites in the primary hypertension group were higher than those in the healthy control group(P < 0.05).The differential methylation sites were mainly distributed in the upstream 2K,3-utr,downstream 2K,5-utr,exons,introns and intergene regions.3.Difference notes: In the result of GO enrichment,the differences biology processes between the two groups were molecular process,single tissue progress,biological correlation regulation,and pathological process.The differenceses of cellular composition were organelles,related tissue functions and related tissue parts in two group.In molecular functions,the two groups were significantly different in function of agglutination-adhesion correlation and catalytic initiation.There were 18 relevant KEGG pathways,and the significant ion pathway was calcium ion pathway.Conclusion:The 6m A difference sites are related with the primary hypertension.
