| Objective: To investigate the relationship between the expression of IL-32 in esophageal squamous cell carcinoma tissues and its curative effect after radiotherapy.According to the conclusions obtained from the collected clinical data,the relationship between the expression level of IL-32 and the radiosensitivity of esophageal squamous cell carcinoma cells was further studied,and the mechanism of its influence on radiosensitivity was revealed.Methods: 1.Retrospective analysis of clinical data of 95 patients with esophageal cancer treated in the Department of Radiotherapy of Lianyungang Dongfang Hospital and Lianyungang Second People’s Hospital from January 2014 to January 2016.Single-and multi-factor analysis Clinical pathology and curative effect.2.Eca-109 and TE-10 esophageal squamous carcinoma cell lines were down-regulated and up-regulated while combined with radiation treatment.CCK8 test was used to detect tumor cell proliferation ability;and plate cloning experiment was used to understand the effect of IL-32 expression level on radiosensitivity of esophageal squamous cell carcinoma;Western bolt test was used to detect STAT3,p-STAT3 and Expression level of apoptotic protein.Results: 1.The difference between the expression of IL-32 and the degree of tumor differentiation was statistically significant(P <0.05).Compared with the low expression group,the IL-32 high expression group had better curative effect and statistical significance after radiotherapy(P <0.05).Combining chemotherapy and high expression of IL-32 are independent factors affecting the prognosis of patients with esophageal squamous cell carcinoma undergoing radiotherapy(both P <0.05).2.Through plate colony formation experiments and CCK8 experiments,the combination of high expression IL-32 group can significantly inhibit the growth of esophageal squamous carcinoma cell lines Eca-109 and TE-10 cells.The high expression of IL-32 plays a synergistic role with irradiation,inhibits the expression of STAT3 and p-STAT3,and increases the expression of the apoptotic protein Bax.Conclusion: The high expression of IL-32 is an independent influencing factor of good prognosis after radiotherapy for medium-term esophageal squamous cell carcinoma.IL-32 may increase the radiosensitivity of esophageal squamous carcinoma cell lines Eca-109 and TE-10 cells through the STAT3 pathway. |
