Objective By the method of discussion,the peripheral blood serum metabolites of TBE patients were detected by liquid-mass spectrometry,and the characteristic metabolites were searched for as potential indicators for early diagnosis and screening of TBE patients.The metabolic pathways of characteristic metabolites were understood.Methods Thirty TBE patients admitted to Inner Mongolia forestry general hospital were selected as the experimental group,and thirty healthy people who underwent physical examination at the same time were used as the normal contrast group.Serum samples were analyzed by UPLC-MS technology.The obtained data were preprocessed and normalized with the Mzmine2.0 software and the orthogonal partial least squares discriminant(OPLS-DA)model was built with the SIMCA16.0 software,and then primary screening of metabolites was conducted in forest encephalitis group and normal control group.The m/z of metabolites was input into HMDB database and the specific metabolites between the two groups were screened by statistical analysis.Finally,the diagnostic efficacy of these characteristic metabolites in TBE was evaluated by drawing the ROC curve and calculating the AUC value of 45 characteristic metabolites(taking AUC > 0.9 as the evaluation index).Metaboanalyst was used to analyze the key metabolic pathways of characteristic metabolites in TBE patients.Results Using UPLC-MS technology and OPLS-DA as the differential model,forty-five metabolites related to the course of TBE were screened by mass spectrometry platform metabolomics.The 45 kinds of characteristic metabolites were mainly based on lipids,including 26 lipids and 1 lipid intermediate metabolites,as well as metabolites such as alkaloids,fatty acids and bile pigments.Compared with the healthy control group,there were 12 metabolites with decreased expression and 33 metabolites with elevated expression in the TBE group.There were 19 kinds of metabolites of AUC > 0.9 were indentified and it has been determined by analysis that Lyso PC(15:0),Lyso PA(24:0/0:0),PA(22:2(13Z,16Z)/18:1(11Z),PE(P-16:0e/0:0),PE(22:6(4Z,7Z,10 Z,13Z,16 Z,19Z),PS(Mono Me(13,5)/Di Me(13,5),PG(18:0/20:3(5Z,8Z,11Z),CL,3-O-Sulfogalactosylceramide)can be used as characteristic metabolites of TBE.The pathways of these characteristic metabolites are sphingolipid metabolism,glycerolipid metabolism,glycosylphosphatidylinositol(GPI)-anchored biosynthesis.Conclusion It is an effective method to detect characteristic serum metabolites in TBE patients by UPLC-MS technology platform.There are 9 metabolites in the serum of TBE patients,which may be the indicators of early diagnosis,screening and detection of the disease.The three pathways of sphingolipid metabolism,glycerolipid metabolism,glycosylphosphatidylinositol(GPI)-anchored biosynthesis lay the foundation for the next study on the relationship between lipids and TBE,and provided serum metabolomics information and a new research approach for the pathogenesis of TBE.
