Pharmacodynamic Effect And Mechanism Study Of Kangfuxin Liquid On Simple Vulvovaginal Candidiasis

Background:Candida vaginitis is a common gynecological disease.Due to the loose fertility policies,oral contraceptives,wearing tight chemical fiber underwear,and non-standard use of antibiotics.The vulvovaginal candida infection of female patients in clinical has been exacerbated.The first-line azole antifungal drugs have been repeatedly used for a long-term,and fungal resistance has become a major obstacle to clinical treatment of fungal infections.Due to the long research time and the huge cost for developing new drugs,traditional Chinese medicine preparations are gradually applied to treat vulvovaginal candida infections.Many studies have shown that traditional Chinese medicine and compound prescriptions have good prospects for the prevention and treatment of vulvovaginal Candidiasis in clinical reseach.Objective:The purpose of this study was to evaluate the efficacy of the traditional Chinese medicine preparation Kangfuxin Liquid on Simple Vulvovaginal Candidiasis(VVC),and to study the mechanism of Kangfuxin Liquid that against Candida albicans in vitro and vivo.Methods:In vitro study: The inhibitory activity of Kangfuxin Liquid against Candida albicans was measured by microdilution method.The inhibitory effect of Kangfuxin Liquid against Candida albicans was dynamically studied by time-kill curve method.In vivo study: The therapeutic effect of Kangfuxin Liquid was evaluated by Simple Vulvovaginal Candidiasis mouse model experiment.Periodic acid-silver metheramine(PASM)was used to analyze the adherence of Candida albicans hyphal to the vaginal mucosa.Immunofluorescent staining was used to detect the defense response of host immune.And the enzyme linked immunosorbent assay(ELISA)was used to determine the changes of inflammatory cytokines in the vaginal mucosa before and after Kangfuxin Liquid treatment.Mechanism study: The inhibitory effect of Kangfuxin Liquid against Candida albicans was evaluated by measuring the hyphal development and aggregation of Candida albicans under different culture conditions in vitro.Results:Kangfuxin Liquid has a good anti-Candida albicans effect in vitro and in vivo.Kangfuxin Liquid can inhibit the hyphal development and aggregation of Candida albicans under different culture conditions in vitro.And it can significantly improve the general condition of mice,increase body weight,relieve vulvar swelling and local inflammation,and reduce the fungal colonization in the vaginal mucosal,Kangfuxin Liquid can clear the colonization of Candida albicans by inhibiting the adherence of Candida albicans hyphal.And it can restore the host immune response and improve local inflammation by improving host Th17 immune function in vivo.Conclusion:Kangfuxin Liquid has significant effect on Simple Vulvovaginal Candidiasis.The main mechanism may be to inhibit the hyphal development,adherence and aggregation of Candida albicans,reduce the colonization of Candida albicans in the vaginal mucosal.And it restores the host immunity,improves the host acquired immune function,clears the colonization of Candida albicans in the vaginal mucosal,relieves local inflammation.
