Backgroud & Aim:Adenine nucleotide translocase 1(ANT1)is a mitochondrial membrane transporter encoded by the gene SLC25A4(solute carrier family 25 member4),which is related to the oxidative phosphorylation metabolic pathway of cancer cells and the opening of the mitochondrial transition pore(MPTP).It can regulate the apoptosis of tumor cells.ANT1 promotes the apoptosis of tumor cells,which has been confirmed by researchers in vitro studies of breast cancer,cervical cancer,pancreatic cancer and other tumors.Colon cancer is the world's fourth largest malignant tumor,in China,the incidence of more people,the 5-year survival rate after the general can reach60%.There is little literature on the expression and role of ANT1 in colon cancer.This study examined the expression of ANT1 in colon cancer and paracancer samples,and analyzed the data and follow-up data of clinical patients,hoping to find the association between ANT1 and colon cancer.Methods:1.Tissue specimens: a total of 76 samples from patients undergoing colon cancer surgery in jiangsu subei people's hospital from June 2012 to June 2013 were collected in this experiment.All samples included cancer and paracancer samples more than 6cm from the cutting edge.All the patients were confirmed to be malignant tumors by pathology after the operation,and the possibility of secondary metastasis was excluded according to the imaging and intraoperative findings.2.Collection of clinical data: we analyzed the data of age,gender,longest tumor diameter,tumor site,tumor differentiation degree,pathological type,TNM stage,and number of lymph node metastasis of all patients with colonic malignant tumor,and followed up 76 patients with survival and recurrence within 60 months by telephone.3.Test methods:We used immunohistochemistry to measure the expression of ANT1 in all colon cancer and matched paracancer samples.SPSS19.0 software was used for data analysis,and the relationship between ANT1 expression and clinical data of patients was analyzed by chi-square test.Kaplan-meier survival function was used to analyze the prognosis of ANT1 and survival and recurrence.Log-rank test P < 0.05 was considered statistically significant.COX risk ratio model was used to analyze the influence of ANT1 and clinicopathological variables on patient survival and recurrence.Results:1.The high expression rate of ANT1 protein in colon cancer samples was 53.94%,and the high expression rate in the matched paracancer tissues was 7.04%,the difference was statistically significant(P<0.05).The expression of ANT1 in colon cancer was closely correlated with TNM stage,tumor differentiation degree and number of lymph node metastasis in tumor patients(P<0.05),and was not significantly correlated with the patient's gender,age,tumor site,tumor longest diameter and pathological type(P>0.05).2.The degree of ANT1 expression in colon cancer is related to the prognosis of patients.Compared with patients with low ANT1 expression,the 60-month survival rate of patients with high ANT1 expression is 34.14%VS60.00%,and the difference is statistically significant(P<0.05).The median survival of patients with high ANT1 expression was 44.0 months,the median survival of patients with low ANT1 expression was more than 60 months,and the survival of patients with high ANT1 expression was significantly worse(P<0.05).3.There was an association between the expression level of ANT1 and the recurrence of colon cancer.The 60-month recurrence rates of the high-expression and low-expression ANT1 groups were 66.66% and 35.29%,respectively,with statistically significant differences(P<0.05).The recurrence time of patients with high expression of ANT1 was significantly earlier than that of patients with low expression of ANT1(P<0.05),indicating that patients with high expression of ANT1 were prone to relapse and had a poor prognosis.4.COX multivariate analysis showed that the expression of ANT1 and the number of lymph node metastases were independent factors in determining the overall survival and prognosis of colon cancer patients,but the expression of ANT1 could not be used as an independent indicator in determining the total recurrence of colon cancer patients.Conclusion:1.The expression of ANT1 in colon cancer was higher than that in paracancer,and the expression of ANT1 was higher in patients with later TNM stage,lower degree of differentiation and more number of lymph node metastasis.2.Patients with high expression of ANT1 showed lower survival time and more relapse,with worse prognosis than patients with low expression.3.Targeting ANT1 may improve the survival and prognosis of patients.
