Objective: To observe the Lingguibafa on time open acupuncture point moxibustion on aging model rats of telomere,telomerase reverse transcriptase(TERT),tumor suppressor gene p53,retinoblastoma(Rb)of protein,protein kinase C(PKC)and protein phosphatase 2A(PP2A)expression and the effect of telomere and cell cycle regulation factor from the Angle discusses the anti-aging mechanism of Lingguibafa,anti-aging Lingguibafa to provide more experimental basis.Methods: A total of 40 male SD rats were randomly divided into normal group,model group,Lingguibafa prevention group and Lingguibafa treatment group,with 10 rats in each group.Except for the normal group,the rats were intraperitoneally injected with D-galactose solution to prepare subacute senescence model for 42 days.During the modeling period,the normal group weighed and grabbed normally,and did not inject any medicine.Lingguibafa prevention group in the modeling at the same time to give Lingguibafa on time open acupuncture point moxibustion,after the end of modeling to continue regular feeding;During the modeling period,no treatment was performed in the Lingguibafa treatment group,on the second day after the treatment,moxibustion was performed by opening acupuncture points on time for a total of 24 days.The rats were killed the day after the experiment,Protein content of TERT,p53 and Rb in rat liver tissues were detected by enzyme-linked immunosorbent assay(ELISA),relative telomere length and gene expressions of TERT,p53 and Rb in rat liver tissues were detected by real-time fluorescent quantitative PCR(RT-PCR),and the expressions of PKC and PP2 A in rat liver tissues were detected by immunofluorescence assay.Results:(1)Compared with normal group,the relative telomere length of model group rats in the liver tissue significantly shortened,the difference was statistically significant(p < 0.01),the Lingguibafa prevention group and Lingguibafa treatment group rats relative telomere length was significantly longer than the model group,the difference was statistically significant(p <0.01).(2)Compared with the normal group,the expression of p53 and Rb in the liver tissue of the rats in the model group was significantly increased(p<0.01).The protein content of the rats in the prevention group and the treatment group was significantly lower than that in the model group(p<0.05),The m RNA expression of p53 and Rb and the protein content of Rb were significantly lower than those of model group.(p<0.01).(3)Compared with the normal group,TERT expression in the liver tissues of the model group was significantly lower(p<0.01),while TERT expression in the Lingguibafa prevention group and Lingguibafa treatment group was significantly higher than that of the model group,with statistically significant difference(p < 0.01).(4)Compared with the normal group,the PKC expression in the liver tissues of the model group was significantly lower(p<0.01);the PKC expression in the Lingguibafa prevention group and Lingguibafa treatment group was significantly higher than that in the model group,with statistically significant difference(p < 0.01).(5)Compared with the normal group,the expression of PP2 A in the liver tissues of the model group was significantly higher(p<0.01);the expression of PP2 A in the Lingguibafa prevention group and Lingguibafa group was significantly lower than that in the model group,with statistically significant difference(p < 0.01).(6)There was no statistically significant difference between the Lingguibafa prevention group and the Lingguibafa treatment group(p>0.05),but the data showed that the prevention group was better than the treatment group.Conclusions:(1)Moxibustion by opening points on time with Lingguibafa can effectively delay the shortening of telomere length in liver tissues of aging rats and maintain the stability of chromosomes;It can effectively increase the expression of TERT in liver tissues of aging rats and activate the activity of telomerase to maintain cell proliferation,delay the process of cell senescence.(2)Moxibustion by opening points on time with Lingguibafa can effectively increase the aging of PKC expression in rat liver tissue,lower aging rat liver tissue in the expression of p53 and Rb,lower aging of PP2 A expression in rat liver tissue,increase cell anti-aging ability,further transmission of control signal aging,make the stagnation of the cell cycle was restored,cell senescence process and achieve the goal of anti-aging.(3)Both the Lingguibafa prevention group and the Lingguibafa treatment group had good effects on delaying senility,and the treatment group could achieve the same effect as the prevention group,but the prevention effect might be better if done early.
