Objective:Based on the parameters of routine clinical experiments to select parameters closely related to aging,and screen further based on the total resolution of population?TRP?when overall considering the dispersion and centralization trend of the entire population.Adopt statistic Z-value to standardize the character parameters to make these various parameters comparable for establishing aging evaluation models.And take use of disease group of chronic obstructive pulmonary disease?COPD?to verify the clinical application of models.Using the blood routine parameters,biochemical and coagulation parameters of intensive care unit?ICU?patients before death to select specific parameters related to death,standardize them with statistic Z-values,and establish the survival advantages to evaluate the survival advantages of severely sick patients that approach death.Based on telomere length?TL?as well as serum biochemical parameters to establish the elder survival advantage model and verified the clinical application of the model within type-2 diabetes mellitus?T2DM?and COPD.And use saliva to select non-invasive aging-related molecular biomarkers,establish differentiating equations of aging evaluation,and use T2DM patients to verify clinical application.Method:1.We selected parameters such as the blood routine,urine routine parameters and serum biochemical parameters of the healthy.Firstly,based on relativity of parameters and ages as well as the sizes of TRP we selected molecular biomarkers of aging characters.Secondly,we standardized the different parameters with statistic Z-values,and established mathematical evaluation model by taking the mean values of various Z index values(Zmean).Finally,we used the receiver operating curve?ROC?to analyzed the discrimination of Zmeanean in the youth as well as elderly groups,and used another healthy group and COPD patients to verify the clinical application of aging evaluation models.2.We selected parameters such as blood routine,serum biochemical parameters and coagulogram near the end-period of ICU patients.Firstly,based on the death time of patients,we selected character parameters of various temporal points during the last month of patients.Secondly,we standardized character parameters with statistic Z-values,and took the mean values of Z-values?ZM?to establish mathematical evaluation models.Finally,we adopted ROC curve to analyze the discrimination of ZM in youth as well as elderly groups,and used models to verify the survival advantages of another dead ICU patients.3.We selected biochemical parameters of the healthy and used qPCR method to examine telomere length of genomic DNA,and selected out survival advantages of aging.We used statistic Z-values to standardize character indicators,established two-dimensional evaluation models of survival advantages and calculated synthetical advantage?ZS?,and used T2DM and COPD diseases to verify the application of models.4.We set T2DM patients and healthy control group,and took the saliva and serum of the same person simultaneously.Firstly,we examined same biochemical parameters of saliva and serum samples.Secondly,we compared relativity of parameters of saliva and serum as well as character of saliva.Finally,based on the correlation of saliva parameters and age groups,we selected out biomarkers related to aging,and established discriminant equations to evaluate aging degree of body.Result:1.Parameters that belong to blood routine,biochemical,urinal or other types and were larger than 0.95 in relation with ages are selected,and classified as urea,glucose and Zmeanean according to TRP from high to low.Zmeanean had a good linear relationship with R2=0.931 and p<0.001 in curve regressions of different age groups.Moreover,in two age groups of 20-30 and over 80-years old,we analyzed Zmeanean values through ROC curves to reach the conclusion that the are under ROC curve?AUC?was 0.982,with p<0.001.Z in both COPD control group as well as ROC curve had an AUC of 0.821.2.Among all the parameters tested,trends of change of 5 parameters including red blood cell count,lymphocyte count,albumin,cholinesterase and B-type natriuretic peptide are closely related to the dying status of patients.Additionally,we evaluated differences of ICU patients 1 day and 30 days prior to death with ZM values of those 5parameters.We analyzed ZM through ROC curves,and can concluded that AUC on ROC curve was 0.828,with p<0.001.ZM was assessed at 1 days before death with comparison to healthy aged people?age>85 years?,and AUC on ROC curve was 0.979,with p<0.001.3.We established two-dimensional evaluation model of survival advantages by respectively transforming the length of albumin and telomere lengths into Z values.There was no significant difference during the evaluation of two-dimensional models of T2MD?p=0.413?,while there was a significant difference in the evaluation of COPD?p=0.002?.When ZS was used to show the overall synthetical advantages of the organism,survival advantages of T2DM was significantly lower than that of the control group?p=0.026?.4.Through comparison as well as analysis of the same parameters of saliva and blood,we selected potassium ions?K+?and lactate dehydrogenase?LDH?in saliva.After taking the logarithm of LDH and K+,we established the discriminant equation?Logistic P?to evaluate the non-aging elderly to an accuracy of 89.50%.The method had an accuracy of diagnosing elderly with an accuracy of 76.70%?p<0.001?.when we evaluated the aging status of diabetes patients and their control groups,there was no significant in the percentages of the aged and non-aged population?p=0.795?;there was significant correlation between Logistic P and age?p<0.001?.Conclusion:1.The three parameters of glucose,albumin and urea Zmeanean were evaluated in youth group as well as the elderly group.There was a significant difference between COPD and its control group,which proved that parameter Zmeanean had potential application values in health selection.2.Overall anaylysis of the 5 parameters of red blood cell count,lymphocyte count,albumin,cholinesterase,and B-type natriuretic peptide ZM values can be used to distinguish differences of the elderly that were 1 days before death and healthy?>85years?,as well as those that were 1 days before death and 30 days before.It also had potential application values in the evaluation of survival advantages of severely sick patients.3.A two-dimensional model of serum albumin and telomere length can be used to evaluate the survival advantage of the elderly.4.The biochemical components in saliva has connections as well as differences with the blood system.Saliva may be a relatively independent system for blood,and LDH and K+had good correlation with age.It can be used as a non-invasive method to assess aging degree after being altered to a discriminant method.
