Objective:To explore the effect of arecoline on the content of Superoxide dismutase(SOD)in serum of SD rats with acute alcoholic myocardial injury,To further explore the mechanism,prevention and treatment of alcoholic myocardial injury,and to provide a new idea for clinical diagnosis and treatment.Methods:Thirty 8-week-old male SD rats were randomly divided into control group,alcohol group and arecoline group(10 rats each).The alcohol group(Group B)was administered with 10 g / kg / d of alcohol diluted with alcohol,and the same amount of saline injection intraperitoneally.The arecoline group(group C)and the alcohol group were administered with an equal amount of alcohol,and 5 mg / kg / d of arecoline was injected intraperitoneally,while the control group(group A)was administered with an equal amount of normal saline,and the same amount of saline injection.The venous blood serum was taken for the detection of various indicators in the three groups of SD rats after 7 days.After anesthesia,the heart specimens were dissected and fixed in10% formalin fixation solution for 24 hours for pathological examination.Serum Myocardial Enzymes were detected by enzyme linked immunosorbent assay(ELISA),the content of Superoxide Dismutase was detected by Xanthine oxidase method,and myocardial pathological changes were observed by HE staining.Biochemical indexes were combined with histological examination,and various methods were used to verify the results of the examination and analyze the dynamic changes of SOD.Result:1.After 7 days of observation,all 10 SD rats in the control group survived,their hair was bright,their activities and reactions were normal,and their spirits were good.All 10 SD rats in the alcohol group and 10 SD rats in the arecoline group survived,their hair was yellow,their activities and responses were slower and their spirits were worse than those of the control group.the body weight there was no significant difference between the control group and the Alcohol Group and arecoline group(P > 0.05),the serum levels of CK,CK-mb and LDH in alcohol group and arecoline group were increased,but the levels of SOD in serum were decreased(P < 0.05),the serum levels of Ck,CK-mb and LDH were increased in alcohol group compared with arecoline group(P < 0.05),The HE staining results showed that the myocardial cells of the control group were arranged neatly,the nucleus was normal and the staining was uniform.Myocardial cells in the alcohol group were disorderly arranged,cell staining was uneven,nucleus degeneration,inflammatory cell infiltration was fine,and vacuoles appeared in cytoplasm.Cardiac cells in the arecoline group were disordered,unevenly stained,and inflammatory cell infiltration were milder than those in the alcohol group.IPP software analysis results: Compared with the control group,the rats in the alcohol group and the arecoline group had significantly reduced myocardial cell counts in myocardial sections and had statistical significance(P <0.05).Compared with the alcohol group,the arecoline groups had significantly higher myocardial cell counts in myocardial tissue sections.Increased,the results were statistically significant(P <0.05).Conclusions: 1.SD rats cause myocardial damage after acute alcoholism,and the serum SOD content decreases.2.Ingestion of arecoline in SD rats may play a protective role in myocardial damage caused by acute alcoholism,and its mechanism may be related to SOD inhibition of oxidative stress.
