Objective: By designing cytology experiments,we intend to enrich the molecular molecular evidence of calcium sulfate(CS)promoting bone formation ability.To explore the effects of calcium sulfate on the proliferation of osteoblast-like MG63 cells and the expression of osteoprotegerin(OPG)and receptor activator of nuclear factor-?? ligand(RANKL)for finding it promotes osteogenic mechanisms to provide in vitro experimental evidence.Methods: 1.Calcium sulfate artificial bone extract was prepared.The cell medium containing calcium sulfate was set as the experimental group,while the cell medium without calcium sulfate was set as the control group.2.The mRNA expression level of OPG and RANKL in MG63 cells was detected by Quantitative Real Time PCR,and the expression products were detected by western-blot,and compared with the experimental group.3.Analyze experimental results,the measurement data is expressed by(?x±s),and the independent sample Student's t testis adopted;when the test level(P <0.05),the difference is statistically significant.Results: 1.The growth of cell lines in the two groups was good.In the CCK-8 proliferation test,the absorbance value of MG63 cells cultured in the cell medium containing the extract of calcium sulfate artificial bone was significantly higher than that of the control group at24 h and 72h(P < 0.05).2.Compared with the control group,MG63 cells in the cell medium containing calcium sulfate artificial bone extract showed significantly higher expression of OPG at mRNA and protein levels,and significantly lower expression of RANKL at mRNA and protein levels,the differences are statistically significant(P < 0.05).Conclusion:Calcium sulfate can promote the proliferation of osteoblasts to a certain extent,promote bone formation,and can promote the expression of OPG and inhibit the expression of RANKL,thereby inhibiting bone resorption through the OPG / RANKL / RANK signaling pathway,indirectly enhancing Bone function.
